Objective: To atserve the effect of The effect of P21 gene and c-fos antisense digoneue leotid on the in timal vascular smooth musclecell hyperplasia of venous antografts. To get to Rnow an ideal genetie therapy of venous autografts.Methods: 20 New Zealand rabbits were selected, which were divided evenly into experimental and control groupat random. The right external jugularveins were antografted into common carotid arteries in the same side. The transplanted veins of experimental group were immersed in the adenovirus-mediated P21 gene solution for 15 minutes just before anastomosis and coated with c-fos antisense olioneucleotide glue gel just after anastomosis, while the control group with nothing. The tramsplanted vascular sample were taken at 2 weeks after operation. The quantity of vascular smooth muscle ceccs (VSMC), expression of proliferating cell nuclear antigen (PCNA),quantity of DNA and the degree of restenosis were investigated.Results: The proliferating index of VSMC of experimental group is 12.74 ± 1.78 and the control group is 26.25 ± 5.75 (P<0.01);Expression of PCNA of experimental group is 32.90 ± 3.64 and the controlgroup is 55.83 ± 3.92 (P<0.01);The quantity of DNA of experimental group is 5.79±1.59 and the control group is 11.62+2.19;The degree of restenosis of experimental group is 11.50±1.85 and the controlgroup is 23.34±5.79 (P<0.01) .Conclusion: ①Fransfection of c-fos antisease oligoneuleotide and P21 gene can infibit the snwoth cells hyperlisia of venous antografts. ②Transfection of c-fos antisense oligoweuleotide and p21 gene is a prospeltive and ideal genetic therapy of restenosis on venous alltografts.
