Aim: To detect differentially expressed proteins for ovarian cancer patient serum.Methods: Surface enhanced laser desorption/ionization time-of-flight mass spectrometry(SELDI-TOF-MS)WCX2 Protein chip technology was used to detect serum samples from 53 patients with ovarian cancer, 50 controls. The resulting profiles between cancer and control were analyzed with Biomarker Wizard Software.Results: 275 protein peaks were detected in the molecular range of 0~50000Da. In total, six protein peaks were significantly detected in sera of patients with ovarian cancer, but not in controls. Among them, five proteins peaks with MW6432, 6879, 8554, 8682, 13726 Da were down-regulated, and one protein with MW11450 Da was up-regulated in ovarian cancer. Comparing the early stage with the late stage ovarian cancer sera, two proteins peaks with MW6190, 6448Da were specificly detected in the early stage ovarian cancer, which were up-regulated. In Swiss-Prot database(http://au.expasy.org/tools/tagident.html), we found 11450Da protein peak accordant with S100 Z protein. S100 Z protein belonged to S100 protein family. Maybe through the interaction with S100P protein, it promoted the development of ovarian cancer. There were no proteins accordant with the other protein peaks, maybe they were new proteins.Conclusion: Differentially expressed proteins were found in ovarian cancer patient serum, which had determinate clinical significance for the early diagnosis of ovarian cancer. SELDI-TOF-MS Protein chip technology is a quick ,easy and practical , high throughput analytic method, and it has wide clinical prospect.
