OBJECTIVE To study the relationgship between the expression of leptin gene in placenta and serum HPL and fetal growth in the patients with hypertension disorder complication pregnancy(HDCP).METHODS The placental expression of leptin gene was determined in 30 women with HDCP(gestational hypertension n=10, mild preeclampsia n=10, severe preeclampsia n=10) and 15 normal pregnant women(control group) by reverse transcription/ polymerase chain reaction (RT/PCR). The serum HPL was measured by immunoradioassay.RESUITS (1) The expression of leptin mRNA in placenta were significantly higher in severe and mild preeclampsia group than that in controlgroup(t=4.97, P<0.01;t=3.12, P<0.01), meanwhile, in the severe preeclampsia was higher than that in the mild (t=2.68, P<0.05).The defferences were not significant between the GH(gestational hypertension) group and the normal group (t=1.94.P>0.05).(2) The serum HPL levels in severe and mild preeclampsia group are significantly lower than that of the normal group (t=7.33, P<0.01;t=5.70, P<0.01);meanwhile, in the severe preeclampsia were lower than that in the mild (t=2.23, P<0.05). The defferences were not significant between the GH(gestational hypertension) group and the normal group (t=0.42,P>0.05). (3) There was a negative correlation between the placental leptin mRNA expression levels and newborn weigh in preeclampsia group (P<0.05), There was a negative correlation between the placental leptin-mRNA expression levels and the serum HPL levels in preeclampsia group.( p<0.05), (4) There was a positive correlation between the placental leptin mRNA expression levels in preeclampsia group and the 24-hour urine protein and MAP(p<0.05).CONCLUSION (1) The placental leptin mRNA expression levels in preeclampsia group increased. (2) The pre-eclampsia women' placental leptin mRNA expression levels correlated with pathogenetic condition. (3) The placental leptin mRNA expression levels in preeclampsia group correlated with fetal growth and development.(4) The pre-eclampsia women' placental leptin mRNA expression levels correlated with women's serum HPL levels...
