IntroductionGlomerular mesangium is composed of glomerular mesangial cell ( GMC ) and extracellular matrix ( ECM). The pathologic change of glomerular mesangia-lis the basis which leads to glomerular sclerosis and renal failure. GMC proliferation (during varies of immune and non -immune injuries) lead to aggradation of big molecular substance on glomerular mesangium and the mechanical obstruction on the glomerular capillaries which reduce the glomerular filtration rate (GFR). How to control the GMC proliferation has great significance in reversion of glomerular injury and chronic change of glomerular disease. CCB's protective effect of kidney is hotspot of research. CCB plays an important role in decreasing the blood pressure and protecting the kidney. The experiment reveals the CCB affection on the GMC proliferation and CCB's protective effect of kidney and its mechanism. A great many of researches reflect varies of factors ( both the promoting and inhibiting factors) affect the GMC proliferation. PDGF promotes the GMC proliferation and IL - 10 inhibits it. So we study the expression of IL - 10 and PDGF in GMC and discuss the CCBs effect on GMC proliferation.ObjectiveObserving the conveying of IL - 10and PDGF on GMC under the different concentration of CCB . So to study Whether CCB can inhibite the GMC proliferation or not.MethodCultrued rat GMC were incubated with different concentration CCB for 24 hours . Then they were dyed with the SABC immunohis to chemistry reagent, IL - 10 and PDGF Ab. Computer image - analyzing system was used to analyze those GMC slices.Results1. The dyed - positive cells of IL - 10 and PDGF show some brown particles.2. For IL - lOgroup, the more [CCB] , the more particles.3. For PDGF group, the more [CCB] , the less particles.4. The average light density of add - drug group^IL - 10 is more than control group notably under same [ CCB ]. ( P < 0.05 ).5. The average light density of add - drug groups PDGF is less than control group notably under same [CCB]. (P<0.05).6. The amout of particles of IL - 10 and PDGF isnt different apparently under the same[CCB]. (P>0.05).7. There is dose - dependent trend of IL - 10 and PDGFs particles under different consistency of the same CCB.Conclusion1. There is inhibition of CCB on the GMC proliferation.2. The inhibitions of the two CCB on GMC arent different apparently.3. There is dose - dependent trend of IL - 10 and PDGFs inhibition on GMC proliferation.4. Mechanism of CCB's inhibition on GMC proliferation maybe connected with stoping the inflow of Ca2 + , so the convey way of internal signal is cut off.
