PrefacePercutaneous transluminal coronary angioplasty is a effective method in therapy coronary heart disease . After operation acute coronary artery obliteration easily occurs , in 6 months rsetenosis also easily occurs, just incidence is as high as 30% - 50% , influenced clinical utilization and the forward curative effect of its treatment. Research shows that restenosis is a vascular reaction characteristic with lumens stenosis and obliteration. The form of restenosis is a complex process, mainly includes;Thrombogenesis and the elastic recovery of blood vessel , the release of biological active factors and the migration and proliferation of the vascular smooth muscle , the form of neogenesis intima and extracellular matrix;reparation of endothelial cell and the remodeling of blood vessel etc. Research also shows that matrixmetaloproteinases( MMPs) and intercellular adhesion molecule(ICAM) have important effect in these process.Peroxisome proliferator - activated receptor gamma (PPAR γ) is a transcription factor belonging to the nuclear hormone receptor gene superfamily. Study discovery, in recent years ,it has the function of protecting blood vessel endothelium , regulating cell factor of inflammation and the expresssion of surface adhesion molecule;inhibiting activation of macrophage and migration and proliferation of the vascular smooth muscle;promoting the conveying of choles-terin. So PPARγ has the effect of protecting blood vessel. Thiazolidinediones(TZDs)is a synthetic ligand of PPAR y. Rosiglitazone(RSG) belongs to TZDs. RSG is a kind of medicine , which can control the blood glucose and fat in type 2 diabetes patients in clinical extensively, have gotten better curative effect. Our research aims at the effects of RSG on restenosis, PPAR 7, MMP - 9 and ICAM -1.Material and MedthodsThe restenosis model was established by injuring the rat abdominal aorta endothelium with balloon catheter. 90 rats were randomly dvide into 3 groups: Namely Rosiglitazone ( RSG) group, false injury group ( F) and balloon injury group ( B). Each group divided into 5 subsets: The rats were killed at days 1,3, 7 ,14and 28 respectively after injury. Observe each group specimen following index: neogenesis intima in different groups by morphologic methods ( Haema-toxylin - Eosin staining ) , the area and thickness were measured and analysed by computer graph analysis system;the expression of MMP - 9 and PPAR y was stain by immunohistochemical staining, calcu - late its average dyeing optical density value;the serum level of matrix metalloprpteinase - 9 ( MMP - 9) and intercellular adhesion molecule - 1 (ICAM - 1) was detected by ELJSA assay. . With SPSS13. 0 statistical software, carry out statistics analysis , Significance was set at P< 0.05.ResultCompared with smooth intima and orderly arranged medial VSMC of aorta in the three groups. Rosiglitazone could decrease the intima proliferation and lumen restenosis compared with ballon injury group, but the lumen area was smaller in ballon injuriy group than in the fasle injury group(P <0. 05);Compared with ballon injury group, the intima area was smaller in RSG group(P <0.05) , but the lumen area was larger than ballon injury group ( P < 0. 05). Comparedthe fasle injury group with the ballon injury group and RSG group, PPAR gamma , MMP - 9expression increases, the expression at the end of 7 days afer endothe-lial injury by balloon is higher than 14 day;The RSG group, 7 day and 14 day PPAR gamma, expression is lower than the same time balloon injury group;The serum level of ICAM - 1 and MMP - 9 was lower in RSG group than that of balloon injury group.Conclusion1. RSG could inhibit the restenosis after balloon injury abdominal aorta in rat .2. RSG may prevent and cure restenosis possibly through suppressing the level of ICAM -1 and MMP -9 after balloon injury.
