In research, macroporous resin was used as sorbent and the preparation procedure of the isoflavones was developed. In order to establish the quantitive analysis and pharmacodynamic methods, some high pure compounds are required. RP-HPLC and HSCCC were used to establish the rapid and high efficiency preparation methods. The quality control of the isoflavones was also studied and genistein was used as the standard to determine the content of the total isoflavones by UV spectra, the two isoflavone aglycones (daidzein and genistein) were used as authentic samples to determine their contents by HPLC. A RP-HPLC method followed liquid-liquid extraction has been developed for the determination of daidzein and genistein in SD rat plasma using quercetin as an internal standard, and then the preclinical pharmacokinetics of the isoflavones was studied.
