ObjectiveTo study the relationship between the expression of EGFR, NM23 and Bcl-2 proteins in hypopharyngeal carcinoma and clinical pathology factors, to investigate its value in clinical application.MethodsWe detected the expressions of EGFR, NM23 and Bcl-2 proteins by inununohistochemistry in the tissues of 29 cases with hypopharyngeal carcinoma and 20 cases with normal mucosal epithelia, and investigated its relationship with the clinical pathology factors of hypopharyngeal carcinoma. The TNM staging of hypopharyngeal carcinoma was suggested according to the stage standard of UICC(1997).Among these cases including stage â… ,2;stage â…¡,3;stage â…¢,6 and stage â…£, 18. 18 were originated from pyriform sinus, 9 from posterior pharyngeal wall and 2 from postcricoid area. Results1 .The positive expression rate of EGFR in hypopharyngeal cancer and in cancer adjacent normal tissues, was 17.2% and 0% respectively,but there was no significantly statistical difference(P=0.0704);The positive expression rate of EGFR in poor differentiation cancer was higher than that in high/moderate differentiation cancer and there existed significantly statistical difference (P=0.0169);The expression of EGFR in the group without lymph node metastasis was significantly lower than that in the groups with unilateral and bilateral lymph node metastasis and there existed statistical difference in different groups(P=0.0242).2. The positive expression rate of NM23 was higher in hypopharyngeal cancer than that in cancer adjacent normal tissues,and there existed a significant statistical difference(P<0.001);The expression of NM23 in cases with lymph node metastasiswere lower than those without,but there was no statistical difference(p=1.0).3.The positive expression rate of Bcl-2 was significantly higher in hypopharyngeal cancer than that in cancer adjacent normal tissue and there existed a significant statistical difference(P<0.001);the expression of Bcl-2 in high differentiation cancer was significantly higher than that in poor differentiation cancer, with significant statistical difference(P=0.0421). Conclusion1. The expression of EFGR in hypopharyngeal cancer was related to the pathological differentiation, the worse differentiation of cancer cell,the higher positive expression rate of EFGR was. The positive rate of EFGR was also correlated with lymph node metastasis: the expression of EFGR in cases with lymph node metastasis were higher than those without.So EFGR may serve as an important marker for assessing malignant behavior of hypopharyngeal cancer.2. The positive expression rate of NM23 was higher in hypopharyngeal cancer than that in cancer adjacenr normal tissues,and there existed a significant statistical difference .The expression of NM23 in cases with lymph node metastasis were lower than those without,but P>0.05.3. The positive expression rate of Bcl-2 was significantly higher in hypopharyngeal cancer than that in cancer adjacent normal tissues and there existed a significant statistical difference .The expression of Bcl-2 was related to the pathological differentiation of hypopharyngeal cancer: the higher the cancer was differentiated, the higher the Bcl-2 expression level was observed,but if Bcl-2 serves as a marker for assessing the biological behavior of hypopharyngeal cancer needing to be studied in the future.
