To clarify the susceptible genes associated with type 2 diabetic nephropathy(DN) in Northern Chinese population, we chosed six polymorphims of candidate genes that have been confirmed to be associated with type 2 diabetic nephropathy in other population. Those six polymorphims of candidate genes were investigated in our sample using methods of The polymerase chain reaction (PCR) and -the restriction fragment length polymorphism (-RFLP) and -the allele peculiar length polymorphism (-APLP)analysis in order to ascertain the association of those six candidate genes with type 2 diabetic nephropathy in Northern Chinese population.The polymorphisms of six susceptible genes include the insert/delete polymophism in intron 16 of Angiotensin converting enzyme (ACE),the Trp64Arg polymophism in beta3 -adrener-gic receptor (ADRB3) gene, the 981 C→T polymorphism of protein tyrosine phosphatase lB(PTPlB),the Ala148Gly ( A→G ) polymorphism in paraoxonase2 (PON2) , the 27bpVNTR polymorphism and the 894G→T polymorphism in endothelial nitric oxide synthase (eNOS).The statistical analysis showed that:1. The Ala148Gly (A→G ) polymorphism of PON2 gene is associated with DN;Significant differences were found in the frequencies of the allele A and G.2. The insert/delete polymophism in the intron 16 of ACE gene showed significant association with DN;The DD genotype had relation with the increased risk of DN. Therefor, in BMI≥25kg/m~2 subgroup and in womensubgroup the difference is very significant.3. The 27bpVNTR polymorphism in eNOS gene were associated with DN;The ba genotype and allele a had relation with the increased risk of DN;genotype bb and allele b had negative relation with the risk of DN.,4. The the 981 C-*T polymorphism of PTP1B may be associated with DN ,allelle T may be the risk factor of DN in norther Chinese.5. The Trp64Arg polymophism of ADRB3 gene were associated with T2DM, c had relation with the decreased risk of T2DM. In men subgroup the TT genotype and allele T > A showed significant association with DN .6. No assiciation were found for the polymorphisms in the genes for the 894 G->T polymorphism in eNOS gene.7. We put the five polymorphisms and the enviromental factors together and compared the power of the association with DN, the result is showed: Duration >PON2> WHR>ACE>Age.
