Objective: To study of the protective effect of total saponins of panax notoginseng(PNS) on gut mucosal barrier in rats with intestinal ischemia-reperfusion injury by animal experiment , and reveal PNS new pharmacological effect and new usage, and provide scientific basis for applied to licnic in future.Materials and Methods: Ischemia-reperfusion(I/R) rat model was established by occluding the superior mesenteric artery(SMA) for 1 hour and followed by reperfusion for 3 hours. Healthy S-D rats were randomly divided into 4 groups.Group A was I/R rats(n=10).Group B was rats by sham-operation(n=10).Group C was rats who were treated with PNS during the fifteen minutes before SMA opened via thigh vein infusion(n=20). Group D was rats who were treated with the same volume saline during the fifteen minutes before SMA opened via thigh vein infusion(n=10). Intestinal impairment was evaluated via mucosal histological changes under light and electron microscope and by mesuring endotoxin concentration in blood plasma and measure ratio of translocation of bacteria in liver spleen and mesenteric lymph nodes(MLN).Results: After ischemia for 1 hour and reperfusion for 3 hours , the levels of endotoxin concentration in blood plasma were found to be significantly lower in PNS treated rats(0. 338 + 0. 061EU/ml) compared with saline treatedrats (0.561 ± 0.043EU/ml, P<0.05) and I/R rats (0.553 + 0. 039EU/ml, P<0. 05). The ratio of bacterial translocation in liver spleen and MLN was found to be significantly lower in PNS treated rats(46. 67%) compared with saline treated rats(76. 66%, P<0. 05) and I/R rats (73. 33%, P<0. 05). Mucosal histological examination under light and electron microscope showen the fabric of intestinal tissue in PNS treated rats were fairly integrity and the mucosal of intestinal tissue were obviously injury in saline treated rats and I/R rats .There was no significantly difference on all indexes between I/R rats with saline treated rats .CONCLUTIONS: This reserch has shown that PNS could obviously protect intestinal mucosal barrier of rat with intestinal ischemia-reperfusion injury by comparison of the levels endotoxin concentration in blood plasma-, the ratio of bacterial translocation and the mucosal histological changes under light and electron microscope. PNS has foreground that it can be applied to clinic patients aimed at protecting intestinal mucosal barrier .
