Objective To investigate expression of tissue transglutaminase (tTG) and its role in carcinogenesis of glioma tumors. To study the role of pl6 protein in glioma development, progression and recurrence.Methods Firstly, tTG was detected by immunohistochemical method in 62 astrocytomas, 18 oligodendrogliomas and 10 normal brain tissue; Secondly, the p16 protein expression was determined in normal cerebral tissue, peri-tumor tissue and different grade glioma by LSABC (labeled streptavidin biotin method) immunohistochemical method, and its distribution areas were measured by image analyzing system; their alternated expression in pre-and post-recurrence of tumors and the correlation with recurrent time were statistically analyzed.Results 1. In glioma tumors , tTG expression was heterogeneous locating in tumor and endothelial cells. Immunoreactivity of tTG protein was significantly different among different grades of astrocytomas. Expression intensity of tTG in glioma was higher than that in benign brain tumors. High expression of tTG contributed around the necrosis foci and apoptotic cells in astrcytomas. 2. The expression of pl6 protein decreased in the order of normal cerebral tissue, peri-tumor tissue and glioma, the distribution areas were the p16 protein expression decreased as the pathological grade increased, the distribution areas of astrocytoma (grade I-II), anaplastic astrocytoma (grade III); The distribution areas of pl6 protein expression decreased in recurrent tumors , specially the p16 protein expression area difference was significant higher in those with increased grade than that in unchanged grade ; The recurrent interval time was negatively correlated with pl6 protein expression level.Conclusions tTG expression contributed to tumor malignant progression in malignant brain tumors. The p16 protein expression is correlated with progression and recurrence of glioma, thus further study on mechanism of its expression alternation will be helpful to the prevention and treatment of glioma recurrence .
