In 1970' ,when American had broken out theLD(Lesionnairs'Diease ),there are many reports about LD in a lot of countrys.In 1982',NanJing approved the LD for the first time.There had many citys researched the legionella in the air-condition cooling tower.the rates were 50%-80%.After the disease of SARS,people have attention the sanitation status of air-condition system. The research had known the bacillus contaminated in the air-condition cooling tower,especially the legionella ,and have collected the data of epidemic. Objective:l.To apart legionella from the the air-condition cooling tower in all kinds of sites,such as hotel,the marketplace,hospital which understand the legionella-contamination to the air-condition cooling tower.Inspect the total of bacteria , the coliform group and feces coliform group to know the ralation between the bacteria .2.Use the PCR to inspect the samples and compare the common method.3.To analyse the legionella gene type and ensure the ralation and variation between heredity .Finally we can know the character of legionella distributing and prevailing.Methods: 1. culture Legionella in the air-condition cooling towers with the couting those in plate and identified with the serum method.2. Inspect the pH, the total of bacteria , the coliform group and fecescoliform group to know the ralation between the bacteria .3.Use two primers 5srRNA and mip to approve the legionella and confirm the sensitivity. Use common and PCR method identify the legionella and compare the two methods to know the merit and disadvantage. 3.Used the AFLP method type the legionella and mensurate the A26o> A280 and caculated concentration of DNA. Analysed the AFLP picture and knew the major legionella.Used the Bionumerics soft to analysed the AFLP picture to observe the resemblance between legionella. Results:There are 31 sites contaminated by legionella in all 55 sites ,such as hotesN marketplace ^ hospital.the rate of contamination was about 56.4%.There 53 samples were separated the legionella in 103 air-condition cooling tower samples,the rate of contamination was about 51.5%.Used the serum method ,we were separated 65 kinds of legionella from air-condition cooling,among those were 54 kinds of legionella which were confirmed legionella pneumophila and 15 were non- legionella pneumophila.3.the sensitivity of two primers inspect the legionella is 5srRNA >20 and mip>205 AUse the common and PCR method inspect the air-cooling tower and compar the rate of two methods.we found the PCR method had a higher inspecting rate.In 103 samples ,there are 94.2% in PCR method and 51.5% in common method.5. Use the AFLP method to type the legionella.The lpl had got 9 kinds of pictures ,and marked AFLPl-9,among these ,the AFLP1 and AFLP5 were major types. Ip2-i4had got 7 kinds of pictures and marked AFLP I -AFLPVD, among these, AFLP III is the major type.The distributing of non- legionella pneumophila was disperse .5.Used the Bionemerics soft to analyse the picture,accord the similar coefficient to differentiate the legionella about 10 groups,and the similar coefficient collected between 70%-100%. Conclusionsiwe got the data about epidemic in the sites of ShenZhencity,and apply the credibility data.The AFLP method can know the gene type and ensure the ralation and variation between heredity .at last can know the character of legionella distributing and prevailing.
