Cytosine arabinoside(Ara-C) is an effective one of chemotherapeutic drugs which cure acute leukemia. The effect and the mechanism of Ara-C treating leukemia are not completely same owing to different doses of Ara-C. Therapeutic effects have improved since therapeutic strategies of different concentrations of Ara-C have been applied. Varied clinical doses of Ara-C are applied, including small dose (20 m g/m2/d), regular dose (100-150mg/m2/d), medium dose (0.5-1.0g/m2/d) and large dose(1.0-3.0g/m2/each ,two each for one day ).In recent 3-5 years, small dose of Ara-C has been applied on treating obstinate leukaemia and leukaemia that cannot be cured by regular therapy ,consolidating complete remission(CR).Large dose of Ara-C are employed on early curing acute lymphoblastic leukemia (ALL) and acute nonlymphoblastic leukemia(ANLL).However, the mechanism of small and large dose of Ara-C treating leukemia has reported little in home and abroad. This paper calculated small, regular and large concentrations of Ara-C according to limber formula:μg/ml= (60XD/5000) X2X103,so small, regular and large blood concentrations were 20, 120 and 1500μg/ml, respectively. The apoptosis of HL60 cells induced by different blood concentrations (20,120 and 1500μg/ml) of Ara-C and the change of bcl-2 expressions in the process were studied in this research .The effects of concurrent use of G-CSF on apoptosis of HL60 cells induced by Ara-C were basically explored.After incubation with different concentrations (20 and 120μg/ml) Ara-C for 24,48and72h, and after HL60 cell treated with 1500μg/ml Ara-C for 6,12 and 24h, HL60 cells morphology were observed under phase contrast microscopy and were observed under light microscope after HE staining. The results showed that the cells treated with Ara-C crimpled and shrank, and their morphology was irregular;but the cells of comparison were of regular and symmetrical morphology. Then the structures of cells were observed by transmission electron microscope. The conclusion indicated that membrane blebbing, chromatin condensation ,crescents formation and apoptotic body were seen; however, these phenomena didn't appear in the cells untreated with Ara-C . |
