Objective:To observe the effect of Chinese herbal GengNian I (GN I ) on osteoporotic ovarietomized(OVX) rats models.Methods:80 Wister female rats about 3 months were randomly divided into 4 groups: GN I group, estrogen group , ovarietomized groups, normal groups,25,15,20,20. On the beginning of the experiment, the later three groups were under the operation of ovarietomy. Up to 5 weeks ,the rats in GN I group and estrogen group were administrated GN I and estrogen respectively from baseline.After 17 weeks , the rats were killed to measure biochemical markers(includingALP.Ca.P.E2),take rats weight and reproductive uterus weight measured,and observe microsopic structure and ultrastruture of the bone and the endometrium.Results: As compared with control group,the weight of the rats in the GN I group and the estrogen group were lower(P<0.05). The uterus weight in the estrogen group and GN I group were higher (P<0.05). Serum levels of ALP were statistically lower in the GN I and estrogen groups than those in the control group.The levels of the serum calcium and phosphate did not show any significant difference among all groups. While GN I group can not raise the E2 significantly(P>0.05). The structure and ultrastucture of bone and uterus showed improved and restored signs to some extend in the GN I and estrogen groups comparing with those in the control group.Conclusion:This study has preliminarily illustrated GN I could enhance bone formation, inhibit bone resorption and basically, it is as effective as estrogen. It has preliminarily demonstrated a good promise of GN I on the treatment of osteoporosis.
