The Experimental Study Of The Effect Of Coicis Semen Extraction Through Acupointal Injection On Regeneration Of Rat's Sciatic Nerve After Injuried

Objective: to observe the therapative effect of coicis semen extraction by acupointal injection on the sciatic nerve regenerationMethod: First the extraction of coicis semen active constituents, using two ways: :water extraction and ethanol extraction, totally 24 active constituents have been extracted; then we have successful cultured the Schwann cell in vitro, and to observe the effect of different active constituent (such as light petroleum group, n-butanol group, NKA60%group, AB860%group, 70% alcohol extract acetoacetate group, 90% alcohol extract acetoacetate group, and 90% alcohol extract 90% ETOH group) on the Schwann cell proliferation and expression of mRNA of NGF and bFGF at the different concentrations of 0.5μg/ml, 1.0μg/ml, 2.0μg/ml and 4.0μg/ml. Finally, we observed the therapy effect of these active constituent by acupointal injection on the sciatic nerve regeneration in vivo.Result: 1. light petroleum group at 2.0μg/ml and 4.0μg/ml concentration point, n-butanol group at 2.0 u g/ml concentration point, NKA60%group at 1.0 u g/ml concentration point , AB860%group at 4.0μg/ml concentration point, 70% alcohol extract acetoacetate group at the 4.0μg/ml concentration point, 90% alcohol extract acetoacetate group at 4.0μg/ml concentration point, and 90% alcohol extract 90% ETOH group at 4.0μg/ml concentration point can obviously promote Schwann cell proliferation2. light petroleum group , NKA60%group and 90% alcohol extract 90% ETOH group can obviously promote the expression of Schwann cell's mRNA of NGF and bFGF, both of genes grow up; n-butanol group can promote the expression of Schwann cell's mRNA of bFGF; 70% alcohol extract acetoacetate group and 90% alcohol extract acetoacetate group can promote the expression of Schwann cell's mRNA of NGF.3. 70% alcohol extract acetoacetate group , Light petroleum group and NKA60% group can obviously promote rat's Sciatic nerve regeneration by acupointal injection. 90% alcohol extract 90% ETOH group also have the same effect by acupointal injection.Conclusion: the active constituents extracted from coicis semen can have the...
