Experimental Study On Anti-Cancer Metastasis Of Tyroserleutide And Its Mechanisms | | Posted on:2005-07-30 | Degree:Master | Type:Thesis | | Country:China | Candidate:J X Hu | Full Text:PDF | | GTID:2144360155959453 | Subject:Immunology | | Abstract/Summary: | PDF Full Text Request | | OBJECTION To research the anti-metastasis effect of Tyserleutide on mice melanoma cell B16 and advanced searches for its possible anti-metastasis mechanisms .METHODS We applied the model of mice melanoma B16 cells aritificial lung metastasis in C57BL/6 mice to examine the effects of Tyroserleutide on anti-metastasis.To investigated Tyroserleutide's effect on the form of B16 cells,the proliferation ability and the clone formation ability of mice melanoma B16 cells cultured in vitro. The artificial basement membrance were set up by matrigel and were used to observe the effect of Tyroserleutide on the adhesion ability of B16 cells. The protein level of LN-R, ICAM-1,E-cadhesion of mice melanamo B16 cells after treated by Tyroserleutide were measured by immunohistochemistry(IHC).RESULTS Tyroserleutide can significantly decrease the lung metastasis of B16 cell in C57BL/6 mice in dosages of 5μg/kg/d-500μg/kg/d,specially the dosage of 50μg/ml can significantly decrease the lung metastasis in the sum total of tumor metastasis lesions by 8.2±2.21, the biger lung metastasis lesions by 1.1± 0.32 and the area of lung metastasis lesion by 46503.13 + 4719.58. Tyroserleutide can significantly inhibit the growth of mice melanoma B16 cells in vitro,the rate of tumor inhibition wasl5-25% in dosages of 1-100μg/ml compared with the control group(P<0.05), inhibition effect of 72hrs was higher than 24hrs,48hrs, specially the dosage of 100μg/ml can significantly inhibit growth of tumor cell by 23.4%. Tyroserleutide could inhibit the clone formation ability of B16 cells, and the inhibition rate of the dosage 100μg/ml by 55%.
