[Objective] To investigate the effect of perfluorohexyloctane(F6H8) to the corneal endothelial cells, anterior chamber angle and retina of the rabbit eyes, and also to evaluate the security of perfluorohexyloctane as silicone oil solvent and intravitreal tamponade.[Methods] Thirty pigmented rabbits were randomized into 3 groups, 10 rabbits in each group. Different quantity of F6H8 were injected into vitreous cavity or anterior chamber in the right eyes: 0.2ml F6H8 as a quantity degree was injected into the anterior chamber in group 1 and into vitreous cavity in group 2; 1.0ml F6H8 was placed into the vitreous cavity after the vitreous was compressed by expanding gas in group 3. All left eyes received the same quantity of NS with same methods served as control. Slit lamp biomicroscopy, indirect ophthalmoscopy and intraocular pressure (IOP) measurement were performed at regular intervals in all eyes pre- and postoperatively. Endothelial cell counts, corneal pachymetry were measured in 4 rabbits of group 1 at 2w, 4w, 8w and 12w after the anterior chamber injection, and electroretinogram(ERG) and fundus fluorescence angiography(FFA) were performed repeatedly in 4 rabbits of group 2 and group 3 at the same time. After the last examination these rabbits were killed and the eyes were enucleated. Other 2 rabbits were killed at 2w, 4w, 8w and the eyes were enucleated. Light microscopy(LM) was performed to study the structure of corneal endothelium, anterior chamber angle and retina.[Results] In group 1, the intraocular pressure had increased postoperatively significantly. The inferior corneal endothelium cell density decreased and the corneal thickness increased to the level of control group.
