Objective To assess the effect of fresh amniotic membrane transplantation (FAMT) on proinflammatory cytokines expression in rat corneal alkali injury, and to investigate the mechanisms of corneal alkali injury, anti-inflammation and modulation immunity of fresh amniotic membrane.Methods Seventy SD rats were divided into three groups. Group A included 32 rats whose right eyes were injured by 0.5mol/L NaOH and were untreated with fresh amniotic membrane transplantation. The rats in group B (n=32) were alkali-burned in right cornea and were treated with fresh amniotic membrane transplantation immediately after burn. Group C consisted six normal rats as the control group. Rats were killed at the 1st,4th,7th ,10th day respectively, after being examined with slitlamp microscopy and graded the corneal opacity, neovascularization (NV) and area of corneal defect.The proinflammatory cytokines IL-1α (including interleukin-1α ), IL-6 (interleukin-6) and IL-8(interleukin-8) expressed in alkali-burned corneas were indentified by Western Blotting, then quantitated using enzyme-linked immunosorbent assay (ELISA), furthermore the mRNA of interleukin-1α were measured by reverse transcription -polymerase chain reaction (RT-PCR).Result At 4,7,and 10 days after the burn, in FAMT-treated corneas, there was less opacification and neovascularization and faster epithelial regeneration, infiltration of polymorphonuclear leukocytes(PMNs) was also alleviated. In normal cornea expression of IL-1α and IL-8 were detected, except that of IL-6. Expression of IL-1α ,IL-6 and IL-8 were strongly induced in the early stages of alkali-burn. IL-1α reached its peak at the 4th day, and IL-6 and IL-8 reached their peak at the 7th day. FAMT significantly suppressed IL-1α , IL-1α mRNA at the first and fourth day (p<0.01), suppressed IL-6,...
