| Objective: To study the difference changes of cellular immune responsebetween the normal pregnancy and hypertension induced by pregnancy (PIH) andthe possible function in the pathogenesis。Methods: Peripheric blood and sera obtained at 20 normal pregnancy and 50PIH who were divided into three groups including mild, moderate and severe. Allthe samples were removed the possible influence factors. T cell subset wereanalysed with an automated flow cytometer and the cytokine of TNF-α,IL-2,IL-6were determined by using enzyme-linked immunoadsorbent assay (ELISA). MAPwas detected by sphygmomanometer.Results: The number of CD3 T cells was not significant difference among allsamples. The number of CD4 T cell of severe group is higher than that of normalcontrol (p<0.05). A decreased content of CD8 T cell was found in PIH women andthe significant difference exists between the normal group and the moderate andsevere group (p<0.05). The changes of CD4 and CD8 T cells lead to an increaseratio of the CD4/CD8 in PIH compare with the normal pregnancy and thesignificant difference exist between normal pregnancy with moderate and severegroups (p<0.05). The serum concentrations of tumor necrosis factor-alpha (TNF-α)were significantly higher in the group of PIH compare with the control group(p<0.05). The serum concentrations of interleukin-2 (IL-2) were significantlyhigher in the moderate and severe group of PIH compare with normal control(p<0.05). The serum concentrations of interleukin-6 (IL-6) were significantlyhigher in the severe group of PIH compare with control group (p<0.05). The ratioof TNF/IL-6 and IL-2/IL-6 did not have difference significant between the controlgroup and the PIH group. Significant positive correlation exist between CD4 T celland the cytokine of TNF-α,IL-2,IL-6 in PIH group (r=0.435,P<0.01;r=0.377,P<0.01;r=0.349,P<0.05). The significantly positive correlation also exist amongthe cytokine in PIH group(r=0.337,P<0.05,r=0.315,P<0.05,r=0.386,P<0.01).The three cytokines have the significantly positive correlation with mean arterialpressure (MAP) in PIH group (r=0.587,p<0.01;r=0.354,p<0.05;r=0.387,p<0.01)。Conclusion: The changes among T lymphocyte subsets in PIH compare withnormal pregnancy maybe reflect the proper number and ratio of T lymphocytesubset are an important role in maintaining the normal pregnancy. Cytokines ofTNF-α,IL-2,IL-6 may be relevant in the pathogenesis of PIH. The research cannot confirm that changes of all these factors are the developing process of thedisease or the cause of the disease. |
