| BACKGROND: Angiogenesis is involved Tumor, Psoriasis, Atherosclerosis, Microangiopathy in Diabetes Mellitus, chronic arthritis, endometriosis, etc; and manifest the behavior of Blood Stasis in CTM. In our previous studies, It was found that HXKSF, a TCM preparation of activating blood circulation and removing blood stasis, could inhibit angiogenesis. OBJECTIVE: Develop compound preparation of HXKS Dripping Pills. METHODS: Adopting orthogonal experimental design, and with total solid matters and ferulic acid for indicatrix, the extraction process and technical condition of preparation was optimized. Macroporous resin absorbent technique was also be investigated with ferulic acid and pharmacological indicatrix. The identification of angelica, chuanxiong rhizome and safflower, and the measurement of ferulic acid in the preparation were be made with TLC and HPLC respectively. Wistar rat were inoculated with sponge at back subcutaneous to establish sponge model. Levels of MVD was observed and compared to study the effect of preparation on angiogenesis. RESULTS: The most proper extraction condition for HXKS Dripping Pill is using 12 times of 80% ethanol, 3 ml/min to diacolation the herbs. D101 macroporous resin could refine the extract. The method of TLC could identify angelica, chuanxiong rhizome and safflower in preparation. In HPLC, the linear range for ferulic acid was 5.2-41.6μg/ml (r=0.9997), The recoveries for it was 100.47% (n=5). The RSD was 1.38% (n=7). In the high-dose and middle-dose HXKSDW treated sponge model rats, MVD was inhibited by 46.7% and 15.33% respectively. CONCLUSIONS: The techniques and methodical control of quality of HXKS Dripping Pills are convenient, feasible, stable and credible. Pharmacodynamics experiment shows that the preparation could inhibit angiogenesis. |
