OBJECTIVE: To explore the effects of PGF1 on the reproduction function of testis following ischemical reperfusion injury and investigate its possible molecular mechanism .METHODS: Completed ischemical reperfusion injury pathological model in SD rats . Thirty rats were randomly divided into three equal groups of ten . A group (sham-operated) B group (testicular torsion/detorsion+PGE,) and C group (testicular torsion/ detorsion + saline). All animals were submitted to unilateral 720° testicular torsion , then detorsion were done in one and a half hour. Thirty days later, all rats were killed .All of the left testis were harvested immediately ,weihted their quality ,calculated their organ coefficient and fixed routinely .Then took out their serum and tested sex hormone . The changes of testis interstitial substance , spermatogenic cells and Sertoli's cells were observed by microscope and transmission electron microscope ,mean while detected diameter of contorted seminiferous tubular through ocular micrometer.RESULTS: (1) In the PGE1 treatment group, the changes of the testis structure were almost the same as those in group A, but the pathological changes of the testis structure were much lighter than those in group C. (2)Under microscope: The pathological changes of group B are almost the same as those in group A ,but are much lighter than those in groupC . 3)The organ coefficient of group B compared with that of group A, there was no significantly difference(p>0. 05). But it was much higher than that of group C(/X0. 05). (4) Seminiferous tubular diameter of group B compared with that of group A, there was no significantly difference (p>0. 05). But it was much higher than that of Compared the group C (p<0. 05). (5) Serum sex hormone : There is not a significantly difference between two groups.CONCLUSION: Testicular function following I/R in rats can be significantly prevented and treated by PGE1... |
