Obiective To investigate correlation between part of HLA class II gene and Gestational diabetes mellitus. To make a comparative study of part HLA class II gene frequencies in the cases of gestational diabetes mellitus and type 2 diabetes mellitus. The correlation between plasma adiponectin and insulin resistance was investigated in order to explore the role of adiopnectin in gestational diabetes mellitus and find out the early index in the natural history of diabetes. Methods The HLA class II gene were detected by polymerase chain reaction-sequence specific primers (PCR-SSP) technique in 48 cases of gestational diabetes mellitus, 48 cases of type 2 diabetes mellitus, and 48 normal pregnancy individuals as the control. Fourty women with gestational diabetes and 40 type 2 diabetic patients and 40 healthy pregnant women were investigated. The serum adiponectin lever was assayed by RIA. Meanwhile, FINS, FPG, TG, TC, HDL-C, LDL-C and BMI were measured, and HOMA-IR index was calculated. Result The frequencies of HLA-DRB1~*0301 were significantly increased in the gestational diabetes mellitus group and type 2 diabetes mellitus group(P=0.025, P=0.006), and the frequencies of HLA-DQAl~*0301 were significantly increased in these two groups(P=0.024, P=0.001). There was no significant difference between the frequencies of part HLA class II gene of gestational diabetes mellitus and type 2 diabetes mellitus. The levels of serum adiponectin both in GDM group type 2 diabetic group were lower then those in control( P <0.01, P <0.01). There was no significant difference between levels of serum adiponectin of Gestational diabetes mellitus and type 2 diabetes mellitus. Multiple linear regression showed that the level of serum adiponectin only negatively related with HOMA-IR index(β =-0.092 , P=0.000). Conclusion The results suggested that there is correlation between HLA class II gene and gestational diabetes mellitus. There is perhaps common mechanism involved in both gestational diabetes mellitus and type 2 diabetes mellitus on partly measured HLA class II allele frequencies.Adiponectin may be a protect factor. Hypoadiponectinemia is greatly correlation with insulin resistance. Hypoadiponectinemia in GDM is concerned with happening of gestational diabetes mellitus and type 2 diabetes mellitus,These data suggest that hypoadiponectinemia in GDM may be an early event in the natural history of type 2 diabetes.
