Objective To realize the level of folic acid in the cells of femalebreast cancer patients and the adults, then separate the lymphocytes of the breast cancer patients and the normal human beings, and were cultured in medium containing different concentration of folic acid in vitro, to determine the lymphocytes' nuclear division index( NDI),and frequency of necrotic and apoptotic cells. That's the cytotoxicity in Human lymphocyte. We can investigate the relationship between the concentration of folic acid and the cytotoxicity of lymphocyte for the breast cancer patients and the normal persons . Methods We use the radioimmunoassay to measure the plasma and red cells concentration of folic acid in the female breast cancer patients and the female adults . We culture the lymphocyte of the breast cancer patients and the normal persons in the medium containing different concentrations of folic acid (30nM,120nM and 240nM) in vitro, and in the situation of deficiency and adequacy folic acid ,we use the comprehensive cytokinesis-block micronucleus assay(CBMN) to measure the effect of the cytotoxicity (NDI, Apo and Nec) Results (1) The concentrations of plasma folate in the breast cancer patients were 8. 9±1. 85 nmol/L, the levels were lower than the concentrations of plasma folate in the normal persons: 12. 3 ± 3. 12 nmol/L, the levels of plasma folate have significantly difference between the female breast cancer patients and the normal persons (P < 0. 01 ) The concentrations of red cells folate in the breast cancer patients were 644. 7 ± 79. 0nmol/L , the levels were lower than the concentrations of red cells folate in the normal persons: 766. 4 ± 109. 1 nmol/L, the levels of red cells folate have significantly difference between the female breast cancer patients and the normal persons ( P<0.01 ) (2) The lymphocytes NDI expectancy in every folate concentration has no significance ( P>0.05) between the breast cancer patients and the normal persons; (3) Respectively cultured by the folate concentration of 30nM,120nM and 240nM; In the condition of 30nM FA the frequency of apoptosis in the lymphocyte of breast cancer patients was higher than that of normal persons ( P<0.01 ); and for the cultured lymphocyte of breast cancer patients, the frequency of apoptosis was significantly higher in 30 nM FA than that in 120nM and 240 nM FA( P<0.01 ), the frequency of apoptosis has no significantly in 120nM and 240 nM FA ( p>0.05 ). (4) The frequency of necrosis in the lymphocytes of breast cancer patients was higher than that of normal persons ( P<0.01 ) ;and for the cultured lymphocyte of breast cancer patients, the frequency of necrosis was significantly higher in 30 nM FA than that in 120nM and 240 nM FA( P<0.01 ), the frequency of apoptosis has no significantly in 120nM and 240 nM FA ( P >0.05 ). Conclusion The plasma and red cells concentration of folic acid in the female breast cancer patients were lower than that in the female adults , the effect of folate deficiency results cytoxicity in lymphocytes of breast cancer patients were harder than that in normal persons, and folate deficiency cause the frequency of apoptosis and necrosis more higher, the adequacy folate have evident genome stability for lymphocyte of breast cancer patients, in the levels of enough folate , we farther increase the concentration of folic acid, can not cause the genome stability in lymphocyte more harder.
