Objective:1. To investigate pretreatment with ibotenate injections into the bilateral pre-and parasubiculum provide protection of the Entorhinal Cortex in coriaria lactone-Induced epileptic rat.2. to study the ethology influence of this pretreatment on coriaria lactone-Induced epileptic rat.3. To evaluate the connection between the Entorhinal cortex and the epileptic ethology of coriaria lactone-Induced epileptic rat.Methods:Ibotenic acid is a useful reagent for making degenerative lesions without prominent local excitation, and enables selective lesioning of the neuronal cell bodies.Pretreated with ibotenate or phosphate buffer Injections into the bilateral Pre- and Parasubiculum ,The rat receive coriaria lactone intramuscular and induced status epilepticus after 5 days. We record the activity of the lactone-Induced epileptic rat bymeans of Digital Video. Twenty-four hours after administration of coriaria lactone , the rat was excecuted respectively. The entorhinal cortex were observated under light microscope After nissl-stained. The high power image were analysised in Image-Pro Plus 4.5 software package. We study the rat activity after administration of coriaria lactone by method of Quantitative ethology analysis. Results:Pretreatment with ibotenate Injections into the bilateral Pre-and Parasubiculum can relieve entorhinal cortex layer III neurodegeneration in coriaria lactone-Induced epileptic rat.The cell area in entorhinal cortex layer III of ibotenate group rats was more large than that of control group rats (P<0.05) . The cell average optical density in entorhinal cortex layer III of ibotenate group rats was more than that of control group rats (P<0.05) . Compared with phosphate buffer group, the rat pretreated with ibotenate display that the initial latent phase of ictal obviously delayed s the duration from the first seizure begin to rat normal activity significantly reduced ^ the persistence time of severe seizure significantly decreased (P<0.05) .Two rats of phosphate buffer group died in two hours after treated with coriaria lactone,but rats of ibotenat group all survival. Conclusions:1. Entorhinal cortex layer III of the lactone-Induced epileptic rat display neurodegenenation.2. Ibotenate injections into the bilateral pre- and parasubiculum ,especial deep layer can help to relieve entorhinal cortex layer III neurodegeneration in coriaria lactone-Induced epileptic rat.3. Ibotenate injections into the bilateral pre- and parasubiculum can partially ameliorate epileptic ethology of coriaria lactone-Induced epileptic rat.4. Entorhinal cortex probabily play an important role in the epileptic ethology amelioratationof the lactone-Induced epileptic rat caused by Ibotenate injections into the bilateral pre- and parasubiculum.Further study of entorhinal cortex will help to know the pathogenesis of epilepsy.
