Objective: To observe the safety of intense pulsed light to fibroblast cell and evaluate the clinic effect of photoaging treated by IPL (Intense PulsedLight)Methods: Chinese hamster lung fibroblast (V79 cell) was chose asexperiment cell and it was passaged and cultured by common method. Cell was gained and inoculated in a twelve-hole culture board at a density of 105cell·ml-1 and one hole inoculate 1ml. The culture board was divide into eight groups including three dosage group of wavelength 560nm and three dosage group of 640nm and one negative control group and one positive group which were irradiation at different dosage of IPL and DNA Ladder test was carried out at once. The MTT value were tested at three time point (24 hour, 48hour and 72hour) and acridine orange staining was carried out at the same time. 33 patients were given out four or more full-face photorejuvenation treated by IPL at an internal of 3-4 weeks. IPL therapy was conducted with the following parameters: wavelength 560nm or 640 nm, energy fiuence of 25-34J/cm2 with pulses of 2.8/6.0ms or 3.2/6.0ms, and interpulse delay time of 20ms. Doctors and patients according to the general appearance of the face and photograph evaluated the effect of IPL and theskin biological parameter such as skin color, skin moisture and skin elasticity were tested by Chromameter CM-2600d, Corneometer, CM825, Cutometer?MPA 580 before every treatment.Results: every dosage group was irradiation at different dosage of EPL appears that no damage happen on DNA that only appears band near the electrophoresis dot and the positive control group appears typical Ladder's band. The outcomes of MTT value suggest the difference of proliferation activity have no statistics significance. The results of acridine Orange staining indicate no obvious apoptosis body formed in every dosage group. The color parameters L* value increased and a* value decreased. The elasticity parameters R2, R5 and R7 increased and the value of skin moisture decreased. The result of visual evaluation by doctors and patients was almost consistent with the skin objective measurements on improvement of telangiectasise and skin texture.Conclusion: PL irradiation is harmless to DNA of V79 cell. The clinic treatments have proved that there were some improvement on skin brightness, telangiectasise and elasticity after treatments and the results by no-invasive biological methods were almost consistent with the visual judgment of doctors and patients. |
