Background—Macrophage-to-foam cell transformation plays an importantrole in the development of atherosclerosis, accumulation of lipid is the maincause. Previous study demonstrated that adiponectin can suppress the class AMSR expression and decreased level of intracellular lipid. To reduce theaccumulated lipid can affect the course of atherosclerosis. The presentstudy investigated the effects of adiponectin on lipid eliminate in humanmonocyte-derived macrophages.Method and Results — Human monocytes were differentiated intomacrophages by incubation in PMA(160nmol/L) for 24 hours. Theadiponectin—treated macrophages contained fewer lipid droplets stained bySudan IV. Adiponectin stimulated the expression of the Bl macrophagesscavenger receptor (SRB1) by immunohistochemical analyses. Treated withphysiological concentration of adiponectin(30ug/mL) reduced intracellularcholesterol ester content, as determined using the colorimetry, enzymaticmethod.Conclusions—The adipocyte-derived plasma protein adiponectin stimulatedeffluxion of intracellular lipid by increasing the expression of SRB1,suggesting that adiponectin may act as not only a preventive but a therapymeasurement to prevent macrophage-to-foam cell transformation inatherosclerosis. |
