| Object To observe the biological response to the wear debris from the total hip prosthesis of Magnesium Silicon Jade (MSJ) in vivo, to compare the capability of inducing osteolysis among the particles of MSJ and the particles of ultra-high molecular weight polyethylene (UHMWPE), and also the particles of alumina ceramic and titanium alloy (Ti6Al4V), which are often used to produce prostheses, and to explore the reasons of loosening of the prostheses of MSJ. Methods MSJ,UHMWPE and alumina ceramic wear debris are produced in vitro, while the wear debris of titanium alloy(Ti6Al4V) is isolated from synovial membrane like interface tissue(SMLIT). 38 Kun Min mice were divided randomly into 5 groups. Three milliliter filtered air was injected subcutaneously on the back of mice, and a further 1ml filtered air was given 2 days later. After 6 days, 3ml(1×10~8/ml) wear debris of MSJ suspensions were injected into established air-pouches in group A(8 mice), while 3ml(1 ×10~8/ml) alumina ceramic particles suspensions in group B(8 mice), 3ml(1×10~8/ml) titanium alloy particles suspensions in group C(6 mice), 3ml(1 ×10~8/ml) UHMWPE particles suspensions in group D and 3ml normal saline was injected in group E as control. Mice were sacrificed 7 days subsequent to particles introduction. One third of each pouch membrane was fixed in 10% formalin for examine by light microscopes, while the remaining tissue was divided into two parts, both were homogenized and centrifuged, the supernanant for detecting the IL-1βand TNF-αby ELISA respectively. Results The content of IL-1βin group A, B, C and D was higher than that in group E,and further, they arranged degressively. As to TNF-α, there was no difference statistically among the whole 5 groups(P>0.05). Conclusion MSJ and UHMWPE particles both can cause histological response and induce the release of inflammatory cytokine -IL-1β, which demonstrated that the wear debris of MSJ was one of the reasons of aseptic loosening of prostheses on the basis of the results of our experiment. Regard to the inductive effect on the osteoclast, out results showed that the particles of MSJ are less active than that of UHMWPE and Ti6Al4V, but more active than that of alumina ceramic. |
