Objective:To investigate the expression of HER-2/neu gene in gastric carcinoma inimmunohistochemistry and illuminate the link between this expression and itsclinical significance and pathology. To analyze the relation between theexpression of HER-2/neu and the survival of gastric carcinoma patients byretrospective study.Methods: 150 patients who had undergone resectable therapyand were approved to be gastric carcinoma by pathology between January 2003 andDecember 2003 were followed-up(end time was January 2005) to make certain howlong the survival was , and to acknowledge the time and cause if the patients hadbeen dead. The experimental group has 80 cases of gastric cancer(age range was30~81 years old, median 55.5, 54 0f man , 26 of woman) who had result offollow-up. The control group consisted of 20 cases with benign gastric diseases. S-Pimmunohistochemisry technique was applied to detect the expression of HER-2/neuin paraffine-embedded specimen. Corresponding clinical data were obtained fromthe medical record department of the first affiliated hospital of JiangXi Medicalcollege. Results:(1). Positive rate of HER-2/neu in the gastric cancer tissue was51.3%, while negative in contrast group. The positive rate of HER-2/neu wasmarkedly higher than the control group(P<0.01)(2).The positive rate of men andwomen in gastric cancer tissue was respectively 55.6% and 42.3%, there was nosignificant difference in sex.(3). Positive rate of Her-2/neu in the gastric cancertissue in ≥60 years old and <60 years old patients was respectively 44.4% and54.7%, with no significant difference(P>0.05).(4).There was also no significantdifference of positive rate of HER-2/neu in the gastric cancer tissue three ofhistological differentiation grades(grade Ⅰ,56.3%;grade Ⅱ,55.0%;gradeⅢ,47.7%)(P>0.05).(5).There was obviously difference of positive rate ofHER-2/neu in the gastric cancer tissue whether there lymphatic and distantmetastasis or not and whether the invasion depth and clinical stages were(P<0.05).(6).The survival of patients with positive HER-2/neu was shorter than those withnegative in terminal gastric cancer(P<0.05).Conclusion:(1).It was demonstratedthat HER-2/neu gene had high expression in gastric cancer tissues. (2).There wasa significant correlation between HER-2/neu positive rate and clinical stagesincluding invasion depth and lymphatic metastasis and distant metastasis , whilethere was no relationship with age, sex, and histological differentiation grades.(3).The survival of patients with positive HER-2/neu was shorter than those withnegative in terminal gastric cancer. (4). The high expression of HER-2/neu is oftencorrelated with more invasive and metastasis ability and also related to shortersurvival and poor prognosis. So HER-2/neu was a good indicator in assessingprognosis of patient with gastric carcinoma.
