Chinese Medicine is lack of clear, objective and internationally accepted standard for raw materials and products, due to its complexity. Its industrialization and internationalization are limited. Hence the study in its quality assessment and control is very essential.In these ten years, fingerprint has become a worldwide-recognized effective technique for Chinese Medicine quality control. Fingerprint mainly uses chromatogram, especially HPLC, as its construction technique, while the study about traditional, cheap and easy TLC is few. Therefore, this thesis tried to use common scanner/digital camera with a digital image processing software to obtain chromatographic fingerprints profiles. The results were checked by CAMAG TLC scanner. The study aimed to find out an appropriate method to assess and control Chinese Medicine quality.In this thesis, Semen cuscutae, Caulis Spatholobi , Fructus Ligustrilucidui , Ginkgoleaves were taken as examples to study fingerprint methodologically as follows:TLC image was obtained by common scanner /digital camera, with digital image processing technique, and was translated into profile. The factors affecting precision and reproducibility were studied. The results obtained by means of image processing technique and CAMAG TLC scanning were compared to find out how similar the results were. The TLCprofile of Fructus Ligustrilucidui obtained under daylight by common scanner was the most similar to that obtained by CAMAG scanner. The TLC profile of Caulis Spatholobi obtained under UV 366nm by digital camera was very close to that obtained by CAMAG scanner, while the fluorescence TLC profile obtained by image processing had more peaks. All the results proved that, image processing method could obtain profiles of TLC, and the results were similar to those got from professional TLC scanner except that some shoulder-peaks were not clear.The system of microemulsion-polyamide film and silica gel plate-organic solvent were compared in the TLC fingerprint analysis of Ginkgoleaves extraction and its injection. The fluorescence intensity of spots were increased and the peaks were more narrow. To select the optimum system of Fructus Ligustrilucidui TLC, uniform design was introduced to arrange the experiment more reasonably and conveniently.The thesis proposed the TLC analysis of extraction of Semen cuscutae, Caulis Spatholobi , Fructus Ligustrilucidui , and Ginkgoleaves and Ginkgoleaves injection, and obtained the fingerprints based on image processing technique. Each fingerprint had more than 10 peaks. The fingerprints of Caulis Spatholobi and Fructus Ligustrilucidui samples' similarity values were above 0. 9, accorded with the Standard of Chinese Medicine Injection Fingerprint Study.The study proved that, the method of constructing TLC fingerprint by image processing has some unique characters: cheap equipment, fast and simple operation, easy preparation of samples, convenience of dealing many samples simultaneously, various information of color/ TLC profiles/similarity values. The proposed method is very fit for GAP production, and can be a reference method in Chinese Medicine quality control.
