Background: The adenine nucleotide translocator (ANT) spans the inner mitochondrial membrane and ANT represents the most prominent Single protein component of the inner mitochondrial membrane . ANT is a bi-functional protein. It facilitates exchange of cytosolic ADP for ATP within the mitochondrial matrix . In addition, as a component of the permeability transition pore, it can dominantly induce apoptosis. Here, ANT can to be a new potential chemotherapeutic target. Objectives: The objectives of our test are as follows: 1, To investigate the dynamical expression of ANT isforms in rats with ischemia /reperfusion. 2, To know the effect of the dynamical expression of ANT on myocardial energetic metabolism in rats with myocardial ischemia and reperfusion; 3, To investigate the relationship between myocardial apoptosis and the dynamical expression of ANT in rats with myocardial ischemia and reperfusion; 4, To explore the changes of myocardial energetic metabolism and myocardial apoptosis with the time of myocardial ischemia-reperfusion . Methods: SD rats, male, were randomly divided into operatedand control groups. The operated group were treated with 20 minutes left anterior descending coronary artery occlusion and 4 hours ,7days reperfusion as ischemia reperfusion model. The relative mRNA content of ANT 1 and ANT2 in myocardium was detected by RT-PCR and gel electrophoresis imaging . Results: 1, The relative mRNA content of ANT 1 got up to the peak At the 4th hour after myocardial ischemia -reperfusion (1.0620 ±0.0151 vs 0.9155±0.0569control, P<0.05) .From the third day after reperfusion, the mRNA content of ANT1 decreased obviously (0.9151 ± 0.0034 vs 1.0620 ± 0.0151 at the 4th hour after reperfusion, P<0.05), there was not significant difference compared with that in control group ( 0.9151±0.0034 vs 0.9155 ± 0.0569 control, P>0.05) . 2, The relative mRNA content of ANT2 got to the maximum on the first day after reperfusion (0.8298 ± 0.0029 vs 0.7599 ± 0.0036 control, P<0.01) . From the third day after reperfusion, the relative mRNA content of ANT2 began to decrease and had not significant statistical difference compare with that in controls(0.7336 ± 0.0028 vs 0.7599 ± 0.0036 control P>0.05) . Conclusions: Both the relative mRNA content of ANT1 and ANT2 got up to the peak within 24th hours after myocardial ischemia -reperfusion.
