Objective: (1) To observe the pathological characters in local foci of patients with cysticercosis; (2) To observe the expression of Thl/Th2 cytokines in local foci of patients with cysticercosis; (3) To detect T-cell subsets and the level of Thl/Th2 intracellular cytokines expression of PMBCs stimulated with cysticercus antigen and onchosphere antigen of Taenia solium in patients and controls; to investigate the role and significance of Thl/Th2 cytokines ( IFN- -γ /IL-4) in immune regulation of cysticercus cellulosae infection .Methods: (1) 45 diseased tissues embedded with paraffin were sliced, we stained the sections with HE, and observed the cysticercus cellulosae morphological feature and the tissues infiltrated with inflammatory cells by the microscope;(2) The IFN- γ /IL-4 expression level in diseased tissues of the patients with cysticercosis was observed by SP immunohistochemical methods;(3) The level of PBMCs T-cell subsets of patients with cysticercosis was detected by flow cytometry;(4) The Thl/Th2 cytokines level of patients with cysticercosis and controls in the PBMCs stimulated with cysticercus antigen and onchosphere antigen of Taenia solium was detected by four-color flow cytometry at 0,5,10,15d; Cellquest or WinMDI2.8 was used in analysis of cytokines and T-cell subsets.Results: (1) There were three histological types in 45 specimens.First, the structure of cysticercus cellulosae of fifteen cases was clear and its bursa walls were thinner, itsinflammatory response was lighter.Second, the structure of cysticercus cellulosae of 17 cases was blurry and degenerate ,the tissue around parasites informed model parasite granuloma, the lining of bursa walls accompanied with mass macrophages, multinuclear giant cells and epithelioid cells , the outer of bursa walls accompanied with lymphocytes ,eosinophils and plasma cells. Third, the parasites of 13 cases were collapse or calcification,the walls around parasites became thicker and accompanied with lymphocytes and plasma cells. (2) The positive rate of IFN- 7 was 77.78%, the positive rate of EL-4 was 53.33%, the positive charcteristic submit buffy granule in the cytoplast of multinuclear giant cells, lymphocytes, macrophages,the positive rate of IL-4 became gradually higher with the increasing of infection time ,IL-4 expression intensity in neurocysticercosis was higher than that of muscle cysticercosis;(3) The percentage of CD3+ and CD4+ T-cells subsets and the ratio of CD4+/CD8 of patients were higher than those of controls in fresh peripheral blood; (4) The percentage of secreting IFN- 7 CD3+of PBMC in patients stimulated with cysticercus cellulosae antigen at 0,5,10,15d was 25.42±5.53%, 30.46±4.94%, 36.52±4.73%, 38.69±5.58% respectively; The percentage of secreting IL-4 CD3+ was 2.52±0.52 %, 3.00±0.57 %, 3.81 ±0.70 %, 5.03±0.73% respectively; (5) The percentage of secreting IFN- 7 /IL-4 CD3+of PBMC in patients fresh blood was higher than that in controls; (6) The level of secreting IFN- 7 / IL-4 CD3+of PBMCs in patients and controls all became gradually higher with the increasing of induction time. (7) The percentage of secreting IL-4 CD4+ cells stimulated with cysticercus cellulosae antigen in controls became gradually higher with the increasing of induction time.Conclusion:(l) The local dieased tissues infected cysticercus cellulosae showed chronic inflammatory and immune reaction and infiltrated with manifold inflammatory cells, to which multinuclear giant cells, lymphocytes and eosinophils gave priority; (2) Thl cytokines secreted by the inflammatory cells were prevalent such as IFN- 7 ,the Th2 cytokines:EL-4 was also present,the positive rate of EL-4 became gradually higher...
