| Lung cancer is one of the most common cancers worldwide and its mortality rate has increased year after year. In China lung cancer has the highest mortality rate among all the cancers for the city drwellers.Despite major advances in lung cancer dignosis and treatment in the past decades,the prognosis of patients with lung cancer has improved only minimally.The overall 5-year survival rate remain less than 15%.If the disease could be diagnosed at the early stage ,the effect of treatment would be improved dramatically and the post-operation survival rate would increase to 38%-55%. Molecular biology has contributed to make people's unstanding to the disease because the gene level and a new idea will be given to the early diagnosis and treatment for lung cancer by the method of molecular biology which can be used to find the association we can adopt beween some gene and lung cancer. p53 gene is the most important cancer-related gene among all the genes.The mutation of some sequence within the p53 may cause the malfunction of p53 protein product and suppress the p53 gene activity .Consequently the cancer will originate and develop.Besides the mutation may also be responsible to decrease the sensibility to radio-and chemotherapy which will cause poor prognosis in the end. SNP(single nucleotide polymorphism), which is a new genetic marker following restiction fragment length polymorism and microsatelite polymorphism , accouts for 80% of all the known polymorhisms.lt has been shown to have the characteristics of high-density inheritance stability and facilitation in analysis automation. As a new method in the genetic study ,SNP almost meet every requiement in the present condition of studies and compensate other study method ,too. Our investigation means to evaluate the association between p53 BstUI SNP and susceptibiliy and radiosensibiliy in NSCLC(non-small cell lung cancer) by case-control test.Blood samples were collected from 50 patients with histological proven diagnosis of NSCLC(IIIB) recruited at the department of radiation oncology , No.2 clinical hospital of Jilin University.Blood sample of 50 healthy donors were collected as control.Then the genome DNA was extract from 5ml blood sample and PCR(polymerase chain reaction) amplification of the fragment involving the p53 BstUI RFLP was performed and the PCR product is 318bp. Following the PCR the PCR product were digested with BstUI at 60°C for 3h.The result of our investigation were analysed statistically and we find that the patients with A1/A2 genotype were sensitive to radiotherapy and the patients with Al/Al or A2/A2 were not sensitive to radiotherapy (x2 =9.2 , PO.05) ,the effective rate to the radiotherapy respectively were 25%,71.4% and 70%. There is no significant relationship between the BstUI SNP in p53 and susceptibility in NSCLC. There are many related reports about association between exon 4 condon 72 polymorphism (BstUI SNP) in p53 gene and susceptibiliy of NSCLC in domestic and oversea studies.However no general conclusion has been reached with respect to the relationship between them.
