Objective: By establishing different animal models, including group I (major lacrimal gland extirpation animal model),group II (major lacrimal gland sclerosing animal model) and group III (experimental serious dry eye syndrome animal model-surgical removal of the lacrimal gland and burning the conjunctival ,meibomain with trichloroacetic), we try to testify the relation between major lacrimal glands damage and xerophthalmia,and to compare differences of these models.To study changes of the different ocular surface tissues,we observe the pathology and ultrastructure of cornea, conjunctiva,meibomain and lacrimal glands of xerophthalmia models. To access the relationship between expression of apoptosis related genes androgen receptor in cornea,conjunctival epithelium cells and meibomain cells of xerophthamia models and ocular surface sicca as well as tissue injury. Material and methods: After producing 10 animals for major lacrimal glands extirpation (group I ) and 10 animals for injecting sclerosant in major lacrimal glands(group II), We made Schirmer test,tear film break-up time(BUT) and fluorescence staining on the eyes at 3 days, 1week, 2weeks ,4 weeks , 8 weeks, 12 weeks after and before the operation,and took 6 normal healthy Wistar mouses as control group. Meantime we produced 10 animals for experimental serious dry eye disease (group III) and above-mentioned examinations were performed on eyes at 3 days, 1 weeks, 2 weeks, 4 weeks and 8 weeks. 5 animals for major lacrimal glands extirpation (groupI) , 5 animals for injecting sclerosant in major lacrimal glands(group II), 5 animals for experimental serious dry eye disease (group III) and 6 normal control groups were obtained.Parts of them were stained by haematine-eosin and observed by microscope.Parts of them were observed by transmit electron microscope. 5 animals for major lacrimal glands extirpation (group I ) , 5 animals for injecting sclerosant in major lacrimal glands(group II),5 animals for experimental serious dry eye disease (group III) and 6 normal control groups were obtained.They were investigated by immunohistochemical staining to detect the expression of Bax and bcl-2. Results :1, Schirmer test and tear film break-up time(BUT) were significantly lower in group III after operation for 3 days and in group I , group II after operation for 1-2 weeks.The difference become more and more prominent in the course of observation (P <0.05), and these conventional tests in group III were all significantly lower (P<0.05) when compared with group I and group II respectively.
