It is a common gynaecological disease which cause is not clear yet .The vascular endothelial growth factor (VEGF) , also known as the vascular permeability factor ( VPF) , is known to be the most specific mitogen for endothelial cells. Hence, it is considered to play a key role in tumor angogenesis. The human hematopoietic progenitor cell antigen(CD34) is reported a sensitive endothelial marker. Staining for CD34 had been used to measure microvessel density (MVD) in tissues. The aim of the present study was to investigate staining patterns of VEGF and microvessel density in the presence of NNEDV,valvar in-traepithelial neoplasia ( VIN) and normal specimen and to detect the relationship between them.MATERIAL AND METHODMaterial; Tissue specimen from patients who had NNEDV and VIN at the First Clinical College of China Medical University from 2000 to 2004 were retrospective examined, including 20 LS, 20 SH, 20 VIN, and 10 NS as control group. All specimen were embeded in paraffin..Experimental apparatus: (omit)Main agents: Monoclonal antibody anti - human VEGF, monoclonal antibody anti - human CD34 and s - p agent box.Method, Principle and Procedure:Method : VEGF and CD34 were stained with S - P irnmunohistochemical method.Principle: According to the principle that antibody can combine with antigen specifically , we use the combination of the biotinglated second antibody , peroxidase and hematoxylin to examine the antigen in the tissues .Procedure : (omit)Contrast: By DAB color, It is positive if the cytoplasma is brown . Counts were carried out in a field measuring at a magnification of 400 x in order to obtain exact quantification of single VEGF - positive cells and microvessels.RESULTVEGF expression: Mean VEGF expression were 9. 50/HP for LS, the differences between LS and NS and between LS and SH were statistically signifi-cant(P<0.05).CD34 expression and MVD; Mean MVD was 10. 10 for LS,the differences between LS and NS and between LS and SH were statistically significant ( P < 0.05). Mean MVD was 32.70 for VIN ,the differences between VIN and NS and between VIN and SH and between VIN and LS were statistically significant (P<0.05).CONCLUSIONStrong VEGF expression for LS ,SH and VIN, also hightest levels of VEGF expression for LS suggested that promoting angiogenesis factors may be involved in LS.The lowest level of MVD expression for LS suggested that microcirculation disorder may be involved in LS.MVD and VEGF exprssion for LS are not consistent, Hence , it more suggested a significant association between LS and microcirculation disorder .
