Objective: The coronary artery disease is a fearful disease that threatens people's life and health. The acute coronary syndromes (ACS) are the serious types of the coronary artery disease which is the important reason of death in elderly patients. Now the study on VEGF has been a focus of attention to prevent and treat ACS. Recently the function of VEGF induced myocardial angiogenesis and the influence of Interleukin-1β(IL-1β) on VEGF has received much attention. In our experimentation, we had three aims: ①To observe whether or not there were differences on the level of VEGF and IL-1βin the serum between the aged ACS patients and aged SAP patients;②To analyze whether or not there were differences on the level of VEGF and IL-1βin the serum between the aged ACS patients and middle-aged ACS patients; ③To analyze whether or not there were differences on the level of VEGF and IL-1βin the serum between the aged ACS patients and healthy middle-aged people. At last, a new method of preventing and treating the ACS could be found by the research on the level of VEGF and IL-1βin the serums of elderly patients. Methods: 60 aged patients of coronary heart disease who were older than 65 years were chosen as objects. Among them 30 SAP patients who had not angina for at least 3 months were selected as group A and 30 ACS patients were selected as group B. 30 middle-aged ACS patients were selected as group C. 30 middle-aged people were selected as group D. The following conditions were excluded: severe hepatic and renal dysfunction, malnutrition, malignant tumor, cerebrovascular disease, peripheral vascular disease, inflammation, connective tissue disease, rheumatic disease, malignant anemia, surgery and traumatism. Blood samples were drawn from the peripheral vein in every patient in 24 hours after admission. And the patient avoided eating food for 8 hours before the blood samples were obtained. 4ml of the blood samples was centrifugated by 3000/minute for 10 minutes and got the serums. the serum samples were frozen and stored at -30℃until they were assayed. The serum levels of VEGF and IL-1βwere determined with ELISA Assay and the reagent boxes were bought from Beijing JingMei Biotechnology Company. All of the operation was strictly done with the guidelines. In the same day, the biochemistry blood samples (4ml) were taken from vein, obtained the serum from the blood and determined items included TC, TG, Glu, et al. Measurement data which was normal distribution and homoscedasticity was expressed by mean value ±standard deviation and statistically analyzed with Software package ofSPSS11.0. ANOVA was used to compare the means of multi-groups. When they were significant, LSD method was used. Chi-square test was used to analyze categorical data. The correlation was analyzed by Pearson method. We take P<0.05 as statistic significance level. Resules:1 Age of group A was 78.43±5.42. Age of group B was 79.27±5.91. Age of group C was 49.90±5.42. Age of group D was 46.73±7.86(Table 1).There was not significant difference of age between group A and group B, P=0.606. There was not significant difference of age between group C and group D, P=0.052. There was significant difference of age between group A and group C, P<0.001. There was significant difference of age between group A and group D, P<0.001. There was significant difference of age between group B and group C, P<0.001. There was significant difference of age between group B and group D, P<0.001 (Table 2). There was not significant difference of other data such as sex, acute myocardial infarction (AMI), diabetes mellitus, blood pressure, TC, TG and treatment. P>0.05 (Table 1). 2 Level of VEGF in group A(96.32±19.97pg/ml) was much lower than that in group B (497.48 ±25.29pg/ml), P<0.001. Level of VEGF in group A(96.32±19.97pg/ml) was much lower than that in group C (515.75 ±25.12pg/ml), P<0.001. Level of VEGF in group A(96.32±19.97pg/ml) was much lower than that in group D (111.11 ±16.63pg/ml), P=0.011. Level of VEGF in group B(497.48±25.29pg/ml) wasmuch lower than that in group C (515.75 ±25.12pg/ml), P=0.002. Level of VEGF in group B(497.48±25.29pg/ml) was much higher than that in group D (111.11 ±16.63pg/ml), P<0.001. Level of VEGF in group C(515.75±25.12pg/ml) was much higher than that in group D (111.11 ±16.63pg/ml), P<0.001(Table 4). 3 Level of IL-1βin group A(9.56±2.22pg/ml) was much lower than that in group B (20.11±3.16pg/ml), P<0.001. Level of IL-1βin group A(9.56±2.22pg/ml) was much lower than that in group C (22.21±3.03pg/ml), P<0.001. There was not significant difference between the level of IL-1βin group A(9.56±2.22pg/ml) and that in group D (9.91±2.26pg/ml), P=0.62. Level of IL-1βin group B(20.11±3.16pg/ml) was lower than that in group C (22.21±3.03pg/ml), P=0.003. Level of IL-1βin group B(20.11±3.16pg/ml) was much higher than that in group D (9.91±2.26pg/ml), P<0.001. Level of IL-1βin group C(22.21±3.03pg/ml) was much higher than that in group D (9.91±2.26pg/ml), P<0.001(Table 5). 4 The correlation was analyzed by Pearson method. The level of VEGF showed positive correlation with level of IL-1βin group B, r=0.707, P<0.001(Fig 3).The level of VEGF showed positive correlation with level of IL-1βin group C, r=0.707, P<0.001(Fig 4). Conclusion: The serum level of VEGF in old ACS patients was higher than that in aged SAP patients and middle-aged people because there was a lack of oxygen and blood in heart of...
