The Changes Of Cytokines On Serum And Lung Tissue In Simulated Weightlessness In Tail-suspended Rats

ObjectiveTo investigate the changes and significines with TNF-a, IL-6, IL-8 and ICAM-1 on the serum and NF-κB, ICAM-1 on the pulmonary tissue in healthy rats.Methods1 .Experiments were conducts on wistar male rats , didived into 4 groups suspended for 7d ,control of 7d and suspended for 21d ,control of 21d .tail-suspended (-30°) was used as weightlessness simulation .2. Tail- suspended ended, to draw the sera and lung tissue from two groups of rats respectively. One group is Tail-suspended for7d and control of 7d, the other group is Tail-suspended for7d and control of 21d. Then according to requirements and to keep them for later test.3. NF-κB and ICAM-1 activity in the lung tissue were detected with the immunohisochemical technique.4. The dynamic variation of sera indicators of tumor necrosis factor a (TNF-a),interleukin-6(IL-6), interleukin-8(IL-8)and intercellular adhesion molecular(ICAM-1) were observed by using ELISA.Results1 The contents of TNF-a, IL-6 and IL-8 in sera of 7d Tail-suspensed rats increased significantly(P<0.01),while the activation level of NF-κB in lung tissue of 7d Tail-suspensed rats increased significantly(P<0.01), moreover those of 21d also rised significantly(P<0.05),but The contents and the activation level of 21d Tail-suspensed rats increased less than that of 7d.
