Objective: To investigate the expression of cyclin E, p2lWAF1/CIP1 and MMP-2 in thyroid carcinoma and its relation to carcinogenesis of thyroid carcinoma. Methods: The expression of cyclin E, p21WAF1/CIP1 and MMP-2 were studied in 47 cases of thyroid carcinoma by immunohistochemical technology . 17 cases of thyroid adenoma, 14 cases of nodular goitre and 12 cases of normal thyroid tissue served as control. Results: (l)In normal thyroid tissue, nodular goitre, thyroid adenoma and thyroid carcinoma, the positive rate of cyclin E protein was 0% (0/12) , 14.3% (2/14) , 47.1% (8/17) , 87.2% (41/47) respectively; p21WAF1/CIP1 protein was 0%(0/12) , 7.1% (1/14) , 29.4% (5/17) , 78.7% (37/47) respectively ; MMP-2 protein was 0% ( 0/12 ) , 7. 1%(1/14) , 23.5% (4/17) , 78.7% (37/47) respectively. The positive rate of three proteins in thyroid carcinoma was notably higher than normal thyroid, nodular goiter and thyroid adenoma( P <0.05). (2) The expression of MMP-2 was correlated with different histologic types of thyroid carcinoma(P <0. 05) ; (3) The expression of MMP-2 was high in thyroid carcinoma with lymph node metastasis, The frequency of overexpression of MMP-2 was associated with lymph node metastasis(P <0.05); (4)There was not a significant correlation betweencyclin E and p21WAF1/CIP1 , cyclin E and MMP-2 protein (P >0. 05). Conclusion: (1) The expression of p21WAF1/CIP1, cyclin E and MMP-2 may play an important role in the development of thyroid carcrinoma; (2)The detection of p21WAF1/CIP1, cyclin E and MMp-2 is valuable todifferentiate between benign and malignant neoplasm of thyroid gland and prognsis.
