Objective:On the basis of past research on the traditional Chinese medicine anti-epilepsy capsule to observe the effect of anti-epilepsy capsule on the pentylenetetrazol (PTZ)-kindled model and study the mechanism of anti-epilepsy capsule at an angle of regulating metabolism of v -aminobutiric acid (GABA). Method:1. The model of pentylenetetrazol (PTZ)-kindled epilepsy was used in the experiment. All rats are randomly divided into five groups:anti-epilepsy capsule I group ? anti-epilepsy capsule II group > sodium valproate group > the PTZ model group, the control group.2. To observe the effect of anti-epilepsy capsule on the weight of the pentylenetetrazol (PTZ)-kindled model.3. To observe the effect of anti-epilepsy capsule on the convulsion of the pentylenetetrazol (PTZ)-kindled model.4. To measured the activity of Y -aminobutiric acid (GABA) -transaminase in homoginized brain tissue of the rats with microultraviolet spectrophotometry.5. To detemine the content of Y -aminobutiric acid (GABA) in homoginized brain tissue of the rats with the high performance liquid chromatography.6. To detemine the content of L- glutamic acid in homoginized brain tissue of the rats with the high performance liquid chromatography.Results:l.At the beginning the weight of the rats of the five groups were not significantly diffirent.But the weight of the anti-epilepsy capsule I group, anti-epilepsy capsule II group> sodium valproate group -. the PTZ model group were significantly different in comparison with the control group (PO.01) . The weight of the anti-epilepsy capsule group I was significantly different in comparison with that of the PTZ model group (P<0.01) .And the weight of the anti-epilepsy capsule I group compared with that of the anti-epilepsy capsule II group was significantly different (P<0.05) . 2.The result of the rate of the anti-epilepsy capsule resisting the model ofpentylenetetrazol (PTZ)-kindled epilepsy:(l)The rate of the anti-epilepsy capsule resisting the model of pentylenetetrazol (PTZ)-kindled epilepsy of the anti-epilepsy capsule I group was significantly increased in comparison with that of the PTZ model group ( P<0.01 ) .And the rate of the anti-epilepsy capsule I group compared with that of the anti-epilepsy capsule II group was significantly diffirent (PO.05) . (2)The convulsion level of the anti-epilepsy capsule group was significantly decreased in comparison with that of the PTZ model group (PO.01) .And the level of the anti-epilepsy capsule I group compared with that of the anti-epilepsy capsule II group was significantly diffirent (P<0.05) . (3) The convulsion latent period of the anti-epilepsy capsule group was significantly increased in comparison with that of the PTZ model group (PO.01) .(4)The convulsion duration of the anti-epilepsy capsule group was significantly decreased in comparison with that of the PTZ model group (PO.01) .And the duration of the anti-epilepsy capsule I group compared with that of the anti-epilepsy capsule II group was significantly diffirent ( P<0.05 ) . 3. The GABA-transaminase activity in the hippocampus and cortex of epilepsy modle group was significantly increased in comparison with that of control group(p<0.01). After treated with the anti-epilepsy capsule the GABA-transaminase activity in the hippocampus and cortex was remarkably decreasd4. The level of the GAB A in the hippocampus and contex of the PTZ model group were significantly decreased compared with that of the control group. After treated with anti-epilepsy capsule the level of the GABA in the Hippocampus and contex of the anti-epilepsy capsule group I was remarkably increased compared with that of the PTZ model group (p<0.01).And the level of the GABA in the hippocampus and contex of the anti-epilepsy capsule II group was remarkably increased compared with that of the anti-epilepsy capsule I group.5.
