In this study, experimentally investigated were the relationship between the proliferative activity of nasopharyngeal carcinoma (NPC) cells and the infection of bacteria and the intervening effects of Five-Ingredient Toxin-Dispersing Beverage (FITDB) on the proliferative activity changes of NPC cells induced by Staphylococcus aureus infection. The experiments were carried out on the infecting model of implanting tumor with NPC cells among nude mice by Staphylococcus aureus in vivo and on the infecting model of NPC cell line HNE3 by the same bacteria in vitro.l.The effects of infection induced by Staphylococcus aureus on the proliferative activity of NPC cells. The infecting model of implanting tumor with NPC cells (HNE3) was prepared by Staphylococcus aureus infection in vivo injected via the tail vein of nude mice following HNE3 cells implanted subcutaneously and implanting tumor grown. Then, the animals were randomly divided into infecting modeling group, treating group with FITDB and controlling group with ampicillin, supplemented with a simply implantingtumor controlling group. They were treated with normal saline, FITDB, ampicillin and normal saline respectively for two weeks. Fourteen days later, the animals were killed to detect the proliferative activity of implanting tumor cells evaluated by proliferative index (PI) based on the determination of PCNA, and then, the results collected in this way were compared each other. The PI of modeling group was significantly increased, accompanied with rapidly increased tumor weight. In contrast, PI of treating group was significantly decreased, with no signicant diference present when compared with that of ampicillin controlling group, all accompanied by tumor weight decreasing. Moreover, NPC cells (HNE3) cultured in vitro also showed significantly increased proliferative activity and significantly higher PI, when infected with Staphylococcus aureus and compared with those cells not infected with bacteria. It was suggested that infection with Staphylococcus aureus should cause very much increased proliferative activity for tumor cells like that of NPC.2.The effects of infection induced by Staphylococcus aureus on apoptosis of NPC cells and the intervening effects of FITDB on this process: Themodeling methods were the same as above. 14 days late following modeling, tissue samples were collected as mentioned above and they were assied with the method of TUNEL to evaluate the changes of apoptotic index (AI). Then, comparison was made among these groups to see the difference of them in the apoptotic activity of tumor cells. The results showed that AI was significantly lower in modeling group than that of other groups, while it was significantly increased when treated with FITDB or ampicillin, accompanied with very lower rapidity of tumor growing. Also, the same condition could be seen in the in vitro experiments on NPC cells. These results indicated that infection of Staphylococcus aureus would cause significantly decreased apoptotic activityfor NPC cells and FITDB could give inducing effects on apoptosis of tumor cells to slow down the growing rapidity of tumor, besides the inhibitory effects on bacteria themselves.
