Objective:To observe the impact of neuroendocrine hormone including adrenine,insulin,noradrenaline and dexasone on the growth and chemotheraphy of pulmonary adenocarcinoma cellA549,to investigate whether the change of sympathetic nerve and endocrine system can influence the growth and chemotheraphy of tumor or not.Furthermore to study whether the influence of those medicine metioned above is dued to affecting the expression of the three protein Bcl-2,p53 and P-Glycoprotein(P-gp).To visit the correlation of the three protein expressed in A549 cell.Method:To place different concentration of adrenine,insulin,noradrenaline and dexasone on A549 cell alone or combinated with xincristine(VCR).(1)MTT assay(mononuclear cell direct cytoxicity assay):to estimate the antirate of those medicine on A549 cell,screening the concentration of IC50 in those medicine.(2)Immunohistology chemistry Technique(IHC):To determinate the influence of those medicine on the expression of Bcl-2,p53 and P-gp ,moreover compare the correlation of the three protein.Result:(1)Different concentration of adrenine,insulin,noradrenaline and dexasone all have effect on the growth of A549 cell is gradually added as also; while the concentration is lowered to one certain level ,even those medicine can accelerate the growth of A549 cell.(2)When those medicine apllied with VCR ,thus can promote the antirate of VCR on A549 cell .(3)The result of IHC show that the combinated application of those medicine and VCR all at IC50 concentration on A549 cell make significantly change of the expression of Bcl-2,p53 and P-gp,the three is all decreased (p<0.01) and the expression of Bcl-2 and P-gp is positively correlated.Conclusion:(1)The result of this article show that in some ranges,different concentration of sympothetic transmitter adrenine,noradrenaline and endocrine hormone dexasone,insulin can have an effect on the growth of tumor in vitro.Along with the adding of concentration ,the inhibition rate is added also;But when the concentration of adrenine,noradrenaline and dexasone is decreased to some level,they can improve the growth of A549.Combined with VCR,the four medicines can enhance the effect of chemotherapy of VCR. .(2)To suspect that change of nervous and endocrine system in vivo may influence the growth of tumor.When the excitability of sympathetic nerve is hightened the growth of tumor will be inhibitted;when the excitability is reduced, the growth will be accelerated.(3)The expression of Bcl-2,p53 and P-gp in VCR bundle is lower than in control(p<0.05),but the expression in associated bundle is significantly lower than in control.The result may be caused by affecting apoptotic gene Bcl-2,p53 and multidrug resistant protein P-gp,leading to the descent of ultraproliferation of A549. In this experiment Bcl-2 and P-gp are positively correlated,which make clear that the yield of multidrug resistance may be involved in the two pathways.
