| Objective: To investigate the role of TGF- β , and TGF- β3 in the occurance and development of abnormal scar and their modulation to Proliferation and apoptosis of fibroblast in abnormal scar which offer the theoretical basis to clinical theropy of abnormal scar .Methods: Tissue specimens which gained from the First Affiliated hospital of Kunming Medical College verified by clinical and pathoological examination during 1997-2003 were obtained from 58 abnormal scars, 42 normal scars and 8 normal skin. The pathological course of scar was 6 month to 2 years which were obtained from the part of head, maxilla, neck et al. 8 normal skin of control group were obtained from head and neck. TGF- β1 , TGF- β3 and PCNA were stained by inmunohistochemistry and terminal deoxynucleotide transferase mediated dUTP nick-end labeling(TUNEL) was used to detect fibroblast' s apoptosis.Results: 1. Inmunohistochemical staining results suggest that the expression of TGF-β, and TGF- β3 was significantly higher in abnormal scar than that of normal scar (P< 0.01). Only one case in 8 normal skin tissues has weak positive expression and the level of its expression was significantly lower than that of abnormal scar and normal scar (P< 0.01). 2. The positive expression of TGF-f3:! was 41.4%(24 of 58 cases) in abnormal scar which was significantly lower than that of normal scar (P<0.01), but in normal skin the expression of TGF- β :, was negative which compared with abnormal and normal scar have significant difference (P<0.01). 3. Compared with normal scar and normal skin the expression of PCNA in fibroblast of abnormal scar increases significantly P<0.01); Compared with normal skin the expression of PCNA in fibroblast of normal scar increases significantly P<0. 05). Compared with normal scar the level of fibroblast apoptosis in abnormal scar decreases significantly (P<0.05); But no significant difference between normal scar and normal skin (P> 0.05). 4. Compare with the relationship between Proliferation Index(PI), Apoptosis Index(AI), TGF-β , and TGF-β , found that there werepositive correlation with the proliferation index and the expression of TGF- P i in fibroblast of abnormal scar(/><0. 05), there were negative correlation with the index of proliferation and the expression of TGF- P :i(P<0. 05) ;On the other hand, there were negative correlation with the index of apoptosis and TGF- 3 , (P<0. 05), no relationship between the index of apoptosis and TGF- 3 :,(f>0.05). Compare with the content between TGF- P , and TGF- P 3 found that there were negative correlation with TGF-P, and TGF-P>Conclusion: 1. TGF-3 is association closely with the occurance of abnormal scar, Increasing expression TGF- P , and/or decreasing expression of TGF- P., may be the one of most important inducible factor. 2. There are interactive relationship in function of the isomer between TGF-P,and TGF-P:) which construct a way of feedback regulation the normal physiological function of fibroblast. 3. TGF- P , and TGF- 3:, regulate the occurance and development of scar by means of regulating the proliferation and/or the level of apoptosis of fibroblast in scar tissue. |
