| Objective:1.To study the secreting level and mRNA expression of the hematopoietic factors IFN % TNF.. IL-3 in the cultured suppernate on the bone marrow mononuclear cells .2. To explore their roles in Kidney yang vacuity type. Kidney yin vacuity type> Dual vacuity of kidney yin and yang type from the chronic aplastic anemia patients.Methods:1. The pareints with chronic aplastic anemia were identified patterns , abstracted 5 ml bone marrow from , and separating bone marrow mononuclear cells, culturing, storing cultured-suppernate and bone marrow mononuclear cells.2. The hematopoietic factors level was measured with ELISA(enzyme-linked immunesorbent assay).3 . mRNA expression of the hematopoietic factors was measured with RT-PCR(reverse transcription polymerase chain reaction).Results:1, The positive IL-3 level of there pattern type groups was significantly lower than control groups(P<0.05). Kidney yin vacuity groups was the lowest, Dual vacuity of kidney yin and yang groups was much lower, Kidney yang vacuity groups was little 1 ower. The positive IL-3 mRNA expression was not measured in any groups.2 ^ The negative IFN- V .TNF- a level of there pattern type groups were higher than control groups(P<0.05). Kidney yin vacuity groups was significantly higher than Kidney yang vacuity groups and Dual vacuity of kidney yin and yang groups(PO.Ol). TNF-a level in Dual vacuity of kidney yin and yang groupswas higher than its in Kidney yang vacuity groups(P<0.05). IFN-. TNFmRNA expression was not measured in control groups.but was measured in there pattern types.Conclusion:The negative IFN- v .. TNF- a level were respectively Kidney yin vacuity type Dual vacuity of kidney yin and yang type. Kidney yang vacuity type from high to low, reversely the positive IL-3 level were respectively Kidney yang vacuity type. Dual vacuity of kidney yin and yang typeN Kidney yin vacuity type from high to low, as can be the foundation to identify patterns for the patients with chronic aplastic anemia. |
