| Purpose: Basic and clinical studies have demonstrated that escin can inhibit inflammation, exudation effects, increase the tension of the vein and ameliorate the micro-circulation of the body. However the pharmacological effects of the elements of escin A,B,C,D have not been reported.We studied the mechanism and the pharmacological effects of the elements of escin A,B,C,D that isolated from escin. Comparing the pharmacological activities which including anti-inflammation, anti- edema, and increasing the tension of the vein, we selected the best composition of escin, which offered a theoretical basis for further developing a new medicine. Materials and Method: Materials: escin and its elements A,B,C,D, inj. Dexamethasone, pertussis bacilli, SD rat, dog, et al. Methods:1. The inhibition effects on the cotton granulomaSterilized cotton 50mg was inoculated in the groin of rats. Drugs were injected into tail vein daily and lasted for 7 days. At the 8th day, the granulomas had been isolated and then baked for 12 hours at 70℃. Then inhibition rates were calculated. At the same time, suprarenal glands and thymus gland of the rats were isolated and weighted. 2.The protective effects on rat infectious brain edema The brain edema model was induced by injection of pertussis bacilli into right internal carotid of rats. Water contents in the brain tissue were measured. And the inhibition rates of different drugs to the rat brain edema were calculated.3. The effects on the vein tensionAfter anesthesia, a part of the dog great saphenous vein was isolated and washed in 4℃ Kreb's-Henseleit buffer, and then cut into 1.5cm pieces. The changes of the vein tension in part of the vein caused by different drugs were recorded.Results:1. The inhibition effects on the cotton granulomaEscin and its elements A,B,C,D can inhibit rat cotton granuloma obviously. The inhibition rates were dose-depended. The elements of secin C,D are better than the others. escin and its elements A,B,C,D could also inhibit rat thymus gland in a dose depended way.2. The protective effects on rat infectious brain edema Escin and its elements A,B,C,D could inhibit rat brain edema caused by pertussis. The protective effects were also dose-depended. Element D was the best one. No statistical significance was found among effects of elements A,B,C,D compared with the group of escin. 3. The effects on the vein tensionEscin and its elements A,B,C,D could induce the contraction of the dog great saphenous vein in vitro. This effect was also dose-depended. No statistical significance was found among effects of elements A,B,C,D compared with the group of escin (p>0.05). Element A had the greatest effect. Conclusion:1. All Escin and its elements A,B,C,D could inhibit inflammation, decrease brain edema and increase the tension of the vein effects in a dose-depended way. The mechanism of anti-inflammation is likely related to the adrenal cortical hormone.2. Evaluation on the experiment of anti-inflammation, the elements of escin C and D had a higher potency and a lower side reaction.3. In anti-brain edema effect, elements of escin A,B,C,D had a higher potency than escin. Among them, element D was the best. |
