| Ulcerative colitis(UC) is familiar intestinal disease. Its pathogenesis is mainly relevant to immunity , heredity , infection and mental nerve . More and more researches have been devoted to studies of UC. Cytokines play a important role in regulating intestinal immunity and are classified into pro inflammatory (IL-1 , TNF- α )and anti-inflammatory (IL-10 ,IL-4). Tumor necrosis factor (TNF)- α is significant inflammation and immunity regulatory factor; Interleukine-10 can regulate activity of T cell and inflammatory reaction; NF- K B is a transcription factor of regulating inflammatory cytokines. In recent years the researches indicate that they can monitor activity of UC . Sulfasalazine , corticosteroids and Immunosu-ppressors are common remedies in treating UC ,but the effects are not satisfied, so exploring mechanism of pathogenesis and pursuing new means of treatment are necessary. Probiotic is microbial medication containing much bacteria. There is evidence that mechanism of treating UC with probiotics are as follows. The first is improving intestinal physical defensive barrier: restoring intestinal perviousness and altering intestinal micro-circumstance and activating generation of benefical bacteria; The second is improving intestinal immunity defensive barrier and eliminating inflammatory reaction. In our study, We observe the results of probiotics on rat with UC by indicating the expression of cytokines then clarify mechanism of UC and find out a new approach of treatment for UC .30 SD rats were divided into five groups: control group, model group, probiotic group, SASP group, combined medication group with SASP and probiotic , there were 6 rats in every group. The normal control group was given 0.9% Nacl by colonic instillation, the other four groups were given 8% acetic acid. Three days later, control and model groups were lavaged by 0.9%Nacl, the three treatment groups were given corresponding drugs by oraladmistration, .After 7 day treatment , The rats in each group were scarificed. Colonic specimens were observed by macrography and dealed with HE staining. The expression of TNF- a NF- K B and IL-10 of the colonic mucosas of rats with UC were detected by a semi-quantitative Immunohistochemistry . The results showed that the three treatment groups not only improve colonic pathologic changes but also decrease the level of TNF-a and NF- K B, increase the level of IL-10 in the colonic mucosas ,the level of TNF- a and NF- K B was significant higher and IL-10 lower than the control group. But the effect of the combined medication was the best among three groups. There was no difference of cytokines in the colonic mucosas between the combined group and the control group.Conclusion: The effect of combined medication is more effective, which is superior to SASP and probiotic group. |
