| Objective Based on establishing Multi-drug resistant strain stably expressing human hepotacarcinoma Bel-7402, whether the CIK induced from PBMC of the HCC (hepatocellular carcinoma) patients possess the cytotoxicy to drug resistant hepatocellular carcinoma cell lines as well as from normal group and its clinical application on hepatocellular carcinoma are observed.Methods 1. Inducing drug resistant Bel-7402 HCC cell line(DR-HCC) by long -term screening of Adriamycin (ADM) culture; 2. Inducing CIK in vitro from PBMC of both normal control group and the HCC patients after operation; 3. Detecting the cytotoxicity of CDC to DR-HCC by MTT method; 4. Based on experimental study, the situation of eleven post-operation patients with hepatocellular carcinoma after the treatment of CIK were observed.Results 1. During CIK culturing, the number CD3+CD56+ cells from HCC patients increased as vigorously as above 600 times. 2. A number of methods demonstrated that MDR1 gene was over-expression and the Pgp function was improvement. 3. In patients group, the cytotoxicy of CIK to the DR-HCC cell lines were about 50%, as same as normal group, and cytotoxicy to L-02 was less then 10%. 4. The result shows that it was safety when the CIK was injected 16 times to 11 patients while no side effects occurring. Both AFP and sICAM-1 level decreased obviously in the patients' serum and 2 patients' AFP mRNA becoming from positive to negative one month after CIK treatment. Tubercular tumor tissue in one liver specimen was observed after CIK treatment, a large amount of lymphocyte cells infiltrated in it , and CD45 , CD3 , CD45RO , CD68 increased greatly by immunohistochemistry.Conclusion The CIK from HCC patients possess all capability as normal group such as strong cytotoxicity to HCC cells and strong cytotoxicity to drug resistant HCC cells in vitro and indeed harmless to normal cells. CIK can offer a safe effective treatment to post-operation patients with hepatocellular carcinoma and offer some help for preventing recurrence as well as metastasis. It should be useful in clinical application and deserve further studies. |
