Objective: Adrenal Medulla Hyperplasia (AMH) is the rare disease in recent 30 years. The pathogenic mechanism of Adrenal Medulla Hyperplasia was not clear. Rescent researsh shows that Tyrosine Hydroxylase (TH) plays an important role in the pathogenic mechanism of Adrenal Medulla Hyperplasia. Now we research on the expression of Tyrosine Hydroxylase gene and TH protein in the adrenals of Adrenal Medulla Hyperplasia Rats, want to find out the molecular mechanism of Adrenal Medulla Hyperplasia.Methods: The Waistar rats were randomly divided into 2 groups: normal control group (NC group), adrenal medulla group (AMH group). AMH models were made by the injection of 0.1% Reserpine 1ml kg-1 d-1, add up to six weeks, normal control groups were injected 0.9% NS 1ml kg-1 d-1, add up to six weeks. At the end of six weeks, adrenals were taken out from all the rats to measure the Tyrosine Hydroxylase protein by immunohistochemical staining and the expression of Tyrosine Hydroxylase mRNA using In Situ Hybridization. At the same time, vein blood was drawn from inferior vena cava to measure the levels of serum NE and E, and 24-hour urine was collected to measure the level of VMA.Results: The expression of Tyrosine Hydroxylase mRNA in group AMH was significantly higher than that in group NC (p<0.01). And the levels of serum NE and E, and 24-hour urine VMA were significantly higher than those in group NC (p<0.01). And TH protein expression had positive corelation with NE, E and VMA (r=0.895, 0.646, 0.667, p<0.01).Conclusion: The expression of Tyrosine Hydroxylase mRNA is increased significantly in adrenals of Adrenal Medulla Hyperplasia Rat. TH expression had positive corelation with NE, E and VMA.TH gene expression too much may be the important reason for the molecular mechanism of Adrenal Medulla Hyperplasia.
