[Objective]: To investigate the effect of DFF45/ICAD and P57KIP2 protein in the carcinogenesis and progression of humam colorectal adenocarcinoma and to identify their correlations with the invasion and metastasis of colorectal adenocarcinoma.[Methods ] : DFF45/ICAD and P57KIP2 protein were detected bySP-immunohistochemistry method in formalin-fixed and paraffin-embeded tissues of 38 normal colorectal mucosal tissues, 31 colorectal adenoma tissues, 45 colorectal adenocarcinoma tissues with no regional lymph nodes metastasis, 38 primary colorectal adenocarcinoma tissues with regional lymph nodes metastasis and their corresponding lymph node metastases.IResults J : The expressions of DFF45/ICAD protein distributed incytoplasms ,it was gradually decreased from normal colorectal mucosal tissues to colorectal adenoma tissues and to colorectal adenocarcinoma tissues (P<0.05); From the well differentiated colorectal adenocarcinoma tissues to the poor differentiated tissues the expressions of DFF45/ICAD protein were significantly decreased (P<0.01) . But the expressions of DFF45/ICAD protein between colorectal adenocarcinoma tissues with and without regional lymph nodes metastasis had no difference (P>0.05) ;the expressions of DFF45/ICAD protein were remain unchanged when primary carcinomas metastasized to lymph nodes (P>0.05). The intensity of the expressions of P57KIP2 protein unchanged fromnormal colorectal mucosal tissues to colorectal adenoma tissues and to colorectal adenocarcinoma tissues (P>0.05). But the expressions of P57KIP2 protein in different lesions distributed in different positions C P<0.01). In normal colorectal mucosal tissues, it mainly distributed in nuclei and cytoplasms ;in colorectal adenoma tissues,it mainly distributed in nuclei and cytoplasms and cell membranes; in colorectal adenocarcinoma tissues, it mainly distributed in cytoplasms and cell membranes and rarely distributed in nuclei ; P57KIP2 expression was not correlated with clinicopathological indices (P>0.05) . There was no obvious correlation between the expressions of DFF45/ICAD and those ofp57KIP2[Conclusions]: The down-regulation of DFF45/ICAD protein and theobstacle of P57 shifting from cytoplasm to nuclei may play important roles in colorectal carcinogenesis and progression.
