Objective To study the bi-directional regulating effect of Puncturervine Caltrop Fruit(PCF) on metabolism of human normal melanocytes(NHM) cultured in vitro and tyrosinase activity by factorial experiment as 3 dosage level and metched with five different Chinese medicine herbs, and try to clarify the mechanism that why it valid in treating different dermatoses of dyspigmentation in clinical, and try to advice doctors use PCF to treat them more efficiently.Methods Culture the human melanocytes in vitro; collect serums from wistar mice after they have been feeded with PCF in 3 dosage(10g, 20g, 30g) metched with five different Chinese medicines or feeded with Methoxsalen(METH) as positive control or water as blank control for 7 days. Determine the content of tyrasinase in serums; Add the serums to the culture medium 3 days, then observ the morphology of NHM, and Counting cells, Hunt method and measuring the rate of oxidation of L-dopa were employed to measure the proliferation, melanin synthesis, tyrasinase activity of melanocytes.Results Compared to blank contrast, PCF and Methoxsalen can improve the content of tyrasinase in blood, and significance of difference was observed (P<0.05) . Different dosages and metchs were found have different effect on melanocytic proliferation, melanin synthesis and tyrasinase activity (P<0.01 ) . Interaction have been found between dosage and metch (P=0.000) . PCF in lOg and 20g dosage can increase tyrasinase activity and depressed NHM proliferation ability; PCF in 30g can decrease tyrasinase activity and increased NHM proliferation ability, and significance of difference was observed (P<0.01) ; PCF metch with Blood-quickening and stasis-transforming herbs or Wind-dissipating herbs can significiently decrease tyrasinase activity. Melanin synthesis decreased as the PCF doseage increased. PCF in 30g and metch with Liver-supplementing and kidney-enriching herbs can significiently decrease melanin synthesis, whilePCF in 10g metch with Wind-dissipating herbs can significiently increase melanin synthesis.Conclusion PCF and Methoxsalen can improve the content of tyrasinase in blood ; PCF affect melanocytic proliferation, melanin synthesis and tyrosinase activity in different dosage or different metched herbs. Low dosage can decrease tyrosinase activity and decrease NHM proliferation, while high dosage can increase tyrosinase activity and NHM proliferation. When use PCF to cure different pigiment disease, in order to improve the effect of cure, we must use the dialectic method with different dosage and metching herbs, such as use high dosage (30g) metch with Liver-supplementing herbs and kidney-enriching herbs to cure disorders of hyperpigmentation such as chloasma, while use low dosage metch Wind-dissipating herbs to cure disorders of hypopigmentation such as vitiligo.
