The methods of treating malignance conventional are operation, radiotherapy and chemotherapy. But all above mentioned can't prevent cancer from metastasis and recurrence. So, recently combination treatment of cancer are emphasized. Immunotherapy is a very important therapy in combination treatment. Within all kinds of methods of tumor immunotherapy, adoptive immunotherapy has become the more important one because of its advantages. This research is going to studying the anti-tumor activity of tumor antigen induced cord blood cells based on the definition of adoptive immunotherapy. 一,Preparing tumor antigen induced cord blood cells Using the method of gradient osmotic pressure and repeatly freezing and melting , tumor antigen was extracted from the cultures lung adenoma (SPC-A-1). Then added the tumor antigen to cord blood cells. After that, the antigen specific tumor killer cells were prepared in vitro. 二,Study the anti-tumor activity of tumor antigen induced cord blood cells (TAK cells) 1,The experiment designs cord blood cells group , cord blood cells plus IL-2 group , cord blood cells plus antigen group , cord blood cells plus antigen and IL-2 group. 2,The non-specific antitumor effect of TAK cells The method of MTT was used to examine the killing activity of TAK cells to K562 cells. Results: Compared with culturing 7 days and 14 days, after culturing 4 days the cells in the cord blood cells plus antigen group and cord blood cells plus antigen and IL-2 group have the strongest killing activity to K562 cells. 3,The specific anti-tumor effect of TAK cells. The method of MTT was used to examine the killings activity of TAK cells to SPC-A-1 cells. Results: Compared with the cells of cord blood cells group, the cells in the cord blood cells plus antigen group and cord blood cells plus antigen and IL-2 groups have the stronger killing activity after culturing 4 days, 7 days and 14 days. 三,Influence of human lung cancer antigen on the apoptosis, cell cycle and cell groups forming of cord blood cells. 1,The experiment designs the groups as above. 2,Results: The apoptosis rate of cord blood cells decreases obviously after induced by tumor antigen and cytokines. The cell division rate of such cells increases obviously and the cell group forming increases obviously, too.
