Ginkgo is special tree in our country. There are more investigations on effects of Ginkgo leaves and its extractions. It was more reported that extract of ginkgo biloba leaves (EGB) could improve learning and memory. But effect of total lactones of ginkgo(TLG)on aging is very few reported. TLG is the important potent component of EGB. The effects of TLG were studied on the natural aging mice model and D-galactose model, and its mechanisms were analyzed. The results as follows:1. Natural aging mice model:TLG could significantly reduce the number of error of learning and memory in step down test. TLG could raise the activities of SOD in blood and cerebrum of aging mice. TLG decreased the content of MDA in cerebrum. TLG could inhibit apoptosis of cerebral cell and decrease the number of apoptotic cells in the brain. The results showed TLG significantly improved learning and memory impairment of aging mice. It may be to enhance the ability of antioxidation and inhibit apoptosis of cerebral cell.2. D- galactose induced aging in mice model:TLG could significantly reduce the number of error of memory and improve the changes of the latent period of the learning and memory in step down test. TLG could raise the activities of SOD in blood and cerebrum and activity of GSH-Px in cerebrum and decrease the content of MDA in blood. TLG could reduce the content of NO anddecrease the activity of MAO in cerebrum. Meanwhile, TLG could reduce the contentof LP in liver and heart and raise the content of Hyp in liver. The results showed TLG significantly inhibit the mice aging induced by D-galactose via attenuating lipid peroxidation and NO.These results showed that TLG possesses the effect on antiaging via attenuating lipid peroxidation and NO and apoptosis of cerebral cells.
