| Objective To study expression of mechanism of three mediums ( substance P, mast cell and histamine ) in abnormal scars and normal skin and the relationship between the mediums and itching or painful of abnormal scarring scars .Methods The immunohistochemistry (IHC) method was applied to study expression of substance P (SP) and mast cell (MC), the fluorometic assay method to study content of histamine (HA)in respective 20 specimens of itching or painful scars and non-itching or painful scars and normal skin tissues;Undegranulated mast cells in the former two groups were counted respectively by microdigital ruler;Semi-quantitative analysis of the IHC results was made by image analysis , using method of t-test, the results were statistically analyzed with the software of SPSS 10. 5 to compare the correlation between the mediums and the abnormal scars.Results 1: In the normal skin and abnormal scars , SP and MC were all positive and HA were all contained. 2. To compare abnormal scars with normal skin, expressions of SP and MC and content of HA (HA : 4. 05+3. 93, 2. 00 + 1. 64 vs 0. 84 + 0. 22) were all significantly higher (p<0.0l) ; 4. In the itching or painful scars, content of HA and expression of SP were all higher than the ones in non-itching or painful scars(p<0.05 in HA;p<0.01 in SP ); MC expression was______________no difference (p>0.05), but the number of undegranulated mast cells was significantly increased(35% vs 23%,P<0.01).Conclusion 1. SP>MC and HA may correlate with the forming of the abnormal scars, higher expressions of SP and MC and higher content of HA may contribute to the scarring of the abnormal scars . 2. SP and HA may correlate with the itching and painful symptom of abnormal scars, higher expression of SP and higher content of HA may induce itching and painful symptom of abnormal scars clinically;Mast cell degranulation plays an important role in the development of the abnormal scars,mast cell causes pain or itch by releasing histamine and other mediators. 3. SP. MC and HA may have some degree relevancewith abnormal scars in its occurrence and development. |
